The astaxanthin(AST)is a kind of carotene with the strongest antioxidant activity,antiseptic and anti-inflammatory activities and also with great research values.However,it is easily inactivated by the influence of light,heat,and oxygen,and the water-insoluble property affects the absorption in the human body.The nano-size embedded astaxanthin will exert great advantages in food,medicine and other industrial fields due to its excellent performance and good sustained release effect.Nano microcapsule technology can improve the water solubility of bioactive substances,enhance the utilization rate of human body and have better sustained release effect.In the present study,poly(lactic-co-glycolic acid,PLGA),Alpha-tocopherol polyethylene glycol 1000 succinate(TPGS)and astaxanthin were used as the wall material,emulsifier and core material respectively.Astaxanthin nano microcapsules in this study was prepared by emulsifying solvent volatilization and ultra-high pressure microjet method.The Characterization analyses and antioxidant activity detection of the nano microcapsules were carried out and the results were as follows:Taking the nano microcapsules size as a major index,the orthogonal optimization of process parameters are: the concentration of TPGS 0.06%(w/v),the concentration of PLGA 20mg/ml,the ratio of water to oil 10:1,the amount of astaxanthin added 1mg/ml,the microjet pressure 15000 psi,2 times the number of micro jets and the stirring speed 2000 r/min.Under these conditions,The embedding rate of nano microcapsule was 93%,the prepared astaxanthin nano microcapsules had the smallest particle size and the nanoemulsion products were the most stable.Nano microcapsules could greatly improve the storage capacity of astaxanthin in normal environment.The particle size distribution of astaxanthin nano microcapsules under the laser particle size analyzer was normally distributed and most of the nanoparticles were distributed within 200 nm.Under the transmission electron microscopy,the nanoparticles were compactly arranged and distributed evenly.In the other hand,the surface was sleek smooth and the particle size was mostly distributed within 100~200 nm.Comparing to the thermogravimetric curves of PLGA,astaxanthin monomer and astaxanthin nano microcapsules,we found that the embedded astaxanthin nano microcapsules in heat resistance increased by 40 ?,and could better protect astaxanthin from degradation.The embedded astaxanthin nano microcapsules exhibited a good slow-release effect compared to common materials,resulted from the change of the polymer material structures according to different purposes.The results of examining the hydroxyl radical and superoxide anion scavenging ability by biochemical methods showed that the scavenging ability of the embedded astaxanthin nano microcapsules was slightly less than the astaxanthin monomer.However,cellular tests showed that the anti-cancer cell activity increased with increasing drug concentration,and the embedded astaxanthin was significantly better than the astaxanthin monomer in anti-cancer activity.
