Preparation,Purification And Microencapsulation Of The Deer Antler Base Polypeptide

Deer antler base is a very hard ossification material that falls off in the following year after a male deer or red deer saws.According to the"Compendium of Materia Medica",deer antler base has reliefed swell and pain,invigorated the spleen,strongthen bones and muscles,promoted blood flow,early treatment of breast milk and other effects.It has a long history for Chinese people to treat mastitis with it,and most studies have shown that the efficacy of the sika antler is closely related to the bioactive peptides.This experiment has conducted a systematic researches on the deer antler base polypeptide separation,purification and and microencapsulation technology,the main conclusions are as follows:1.By utilizing ultrafiltration separated enzynolysis of the deer antler base polypeptide in the test,the best separate condition was:materical-liquid ratio 1.44%,operating pressure0.13 MPa,operating time 60 min.Under this condition,the infiltration flux was 1.72 L/m~2·h,and the content of the polypeptide in the separated sample was 45.2%.2.Utilize the ion exchange column purified the deer antler base polypeptide in the test.Test result showed that the deer antler base polypeptide solution was separated by DEAE-52column,which was eluted with distilled water,0.1 mol/L NaCl,0.2 mol/L NaCl and 0.3 mol/L NaCl,4 components were gained.The antioxidant test showed that the activity of second components was higher,the inhibition rate of hydroxyl radical(·OH)was 69.7%,and the content of polypeptide was 60.3%.3.Utilize the gel column purified the deer antler base polypeptide in the test.Test result showed that the deer antler base polypeptide solution was separated by Sephadex G-50column,buffer with 0.5mmol/L NaCl liquid,2 component were gained.The antioxidant test showed that the activity of second components was higher,the inhibition rate of hydroxyl radical(·OH)was 75.3%,and the content of polypeptide was 73.4%.4.The purified deer antler base polypeptide were analyzed and found it contained 17kinds of amino acids,including 7 kinds of human essential amino acids.The higher contents included leucine,lysine,phenylalanine and valine.HPLC-MS/MS showed that the molecular weight was 715.3224 Da and the peptide sequence was Met-Gln-His-Arg-Tyr.5.In the experiment,microencapsulation technology was used to embed the deer antler base polypeptide,The best process conditions of chitosan embedding the deer antler base polypeptide:wall material:core material was 4,ultrasonic temperature was 30℃,the ultrasonic power was 70W.Under this condition,the embedding rate reached 88.73%.The best process conditions of glutin embedding the deer antler base polypeptide:wall material:core material was 6,ultrasonic temperature was 30℃,the ultrasonic power was 80 W.Under this condition,the embedding rate reached 74.86%.Compared two different wall materials of microcapsules,with gelatin as the wall material of microcapsule had good solubility,can be dissolved in 40℃warm water slowly,solution for light yellow and slightly gamey smell;And chitosan as wall material of microcapsules in 60℃can be dissolved in water,solution for colorless and odorless.The microencapsulated deer antler base polypeptide was not easy to absorb moisture and had good antioxidant activity.
