Osteoarthritis(OA)is a degenerative joint disease,It is characterized by chronic pain and movement disorders in the joints.Although the underlying mechanism of OA remains poorly understood,inflammatory responses and oxidative stress have been proposed to contribute significantly to the initiation and development of OA.Proinflammatory cytokine interleukin-1?(IL-1?)plays a crucial role in the pathogenesis of OA by stimulating several mediators contributed to cartilage degradation.The articular chondrocyte play an important role in maintaining the synthesis of normal tissues and aging extracellular matrix(ECM).IL-1?can induce cartilage degradation by upregulating the expression of matrix metalloproteinases(MMPs),IL-1? can result in the production of reactive oxygen species(ROS)and increased release of inflammatory mediators such as nitric oxide(NO)and increased expression of nitric oxide synthase,It can also promote chondrocyte apoptosis by damaging mitochondrial DNA and inhibiting transcription.Thus,drug of targeting IL-1?-induced inflammation may prove effective as a OA therapeutic strategy.Velvet antler polypeptide(VAP)is a peptide active monomer isolated from velvet antler and has important research value.Existing studies have shown its role in inhibiting inflammation.Therefore,The research focused on the extracted rabbit chondrocytes,was undertaken to evaluate the influence and potential molecular mechanism of VAP attenuates Interleukin-1? induced oxidative stress,apoptosis and inflammatory response in chondrocytes.The VAP used in the experiment was extracted in the early stage of the laboratory and its amino acid sequence was VLSATDKTNVLAAWGKVGGNAPAFGAEALERM.In addition,the animal model of Osteoarthritis was established through the modified Hulth method,and explore the anti-inflammatory effect of traditional Chinese medicine compound velvet antler tonifying kidney soup with special formula.The results showed that VAP can effectively promote the proliferation of chondrocytes within a certain range.VAP effectively blocked the generation of ROS in IL-1?-treated chondrocytes,indicating that VAP has a potent antioxidant activity.VAP attenuated IL-1?-induced production of NO,major inflammatory mediators in these cells,which was associated with the inhibition of inducible iNOS expression.VAP also inhibits IL-1?-induced chondrocyte apoptosis.Moreover,VAP significantly downregulated the level of protein expression of matrix metalloproteinase(MMP)-2,-9 and-13 in chondrocytes treated with IL-1?,resulting in their extracellular secretion reduction.VAP restored the expression of Smad2/3 and P-Smad2/3 by IL-1?-induced activation.These results indicate that VAP may exhibit anti-oxidant,anti-apoptotic and anti-inflammatory effects by interfering with the TGF-?/Smad signaling pathway.Overall,VAP may be a potential effective monomer for the management of OA.The study found that in the rabbit osteoarthritis model and velvet antler tonifying kidney soup gavage treatment,the disorder of white blood cell system occurs in normal body and the velvet antler tonifying kidney soup has a tendency to correct the disorder to a certain extent.Therefore,the effect of velvet antler tonifying kidney soup on osteoarthritis needs to be further deepened to provide more experimental and theoretical basis for the treatment of osteoarthritis with traditional Chinese medicine.
