Collagen is a hydrolyzate with a stable triple helix structure and relatively small molecular weight which is degraded faster and absorbed easier by the organism. It has been reported recently that collagen is widely used in the domain of food, cosmetics, pharmaceuticals and so on owing to its biochemical properties and pharmacological actions.In this paper, the basic components of the antler bas, the use of biological enzyme technology, the deer proteins into antler bas was studied. Extraction of Collagen Extracted of Antler base by pepsin and the collagen powder was frozed by freeze-dried, useing the antler bas peptide and powder to do the anti-fatigue experiments of m antler bas ouse. The main results are as follows:1. Using the national standard for the basic ingredients of the composition of antler bas were detected and analyzed. The results were as follows:ash 58.14%, fat 2.57%, total sugar 1.74%, protein33.27%.2. Using vacuum freeze-drying method and pepsin to make the bas ouse powder, for the single-factor test analysis to determine the best peptides concentration is 10 mg/mL3. Use alkaline protease to hydrolysis antler bas protein to get polypeptide. Apply the response surface methodology to select these four main factors:temperature, time, pH, and enzyme quantity. Regard the hydrolysis rate as the response value to make its process optimize. We obtained the optimum hydrolysis conditions for proteolysis of antler bas: temperature (53%), time (5.5h), pH (9.5), enzyme quantity (1272u/g). In this condition, the actual hydrolysis rate of antler bas was (22.13%). Experiments show that optimization conditions for preparing the deer blood polypeptide using response surface method is feasible, the process parameters we got also have practical value.4. Use membrane filtration and gel chromatography to purify enzymatic hydrolysis substances. Select micro filtration (0.45) and ultrafiltration (10KD) to filtrate hydrolysis liquid, and harvest the gel-filtration liquid. The results show that membrane has separation effects for the hydrolysis substances and removes impurities effectively, which obtain the substance below molecular weight of 10000D. Through UV whole wavelength scan, it was determined that the sample had maximum absorption peak when the time of λ 220nm. Using MALDI-TOF/TOF-MS as measured antlers disc purified polypeptide having a molecular weight 1342.9m/z, and by ESI-Q-TOF-MS to the verified and found 1226.7,1069.8 1010.8m/z and subsequent polypeptide fragments and found that this peptide fragment corresponding to an amino acid fragment of glycine and proline (Gly, pro) which can further determine the hydrolysates from collagen peptide substances. But which also contains specific amino acids, amino acids connection between its yet to be explored.
