| In recent years,the environmental pollution problems of Chinese medicinal materials emerge in an endless stream.The Chinese medicinal materials exported in China have been sealed up many times in the international market because of pollution problems.Pesticide residues and heavy metal pollution have become a "stumbling blocks" that Chinese medicine marches into the international market.Therefore,the content of heavy metals and pesticide residues in Chinese medicinal materials need to be determined and limited,which is the key to ensure the safety of drugs and promote the internationalization of Chinese medicine.Velvet antler as a precious traditional Chinese medicinal material,its pharmacological effect is lasting.For a long time,the research and development of velvet antler has been paid attention to by many experts and scholars,but at present,most of the research on velvet antler is focused on the analysis of its chemical composition,however,the numerous pharmacological effects of it,such as anti-tumor,need to be developed and used.Taking Chinese medicinal velvet antler as the research object,the heavy metal and pesticide residues in nine kinds of velvet antler samples from different habitats,different species and different parts were analyzed,and the pollution of them were evaluated according to the relevant national standards.Next,based on the anti-tumor effect of velvet antler,we explored the effects of heavy metals and pesticide residues on the anti-breast cancer activity of its active ingredients.The contents of 4 isomers of BHCs,4 isomers of DDTs and the content of pentaploron in nine kinds of velvet antlers were detected by gas chromatography,nine kinds of velvet antlers are New Zealand Sika Deer powder,red powder,wax slice,Sika Deer(Jilin)red powder,yellow powder,wax film,Wapiti(Jilin)Flakes of powder,sand and wax.The contents of Pb,Cd,Cu,As and Hg in nine kinds of velvet antlers were detected by ICP-MS.The results showed that the content of these harmful substances in the nine kinds of velvet antler samples were all in accordance with the national limit standard.However,this study found that there was a certain amount of pesticide residues in the velvet antler as an animal medicine,and some indexes,such as heavy metal copper,were high,which suggests that we need to detect and analyze heavy metals and pesticide residues in velvet antler before using them so as to ensure the safety of people’s medication.In this paper,the New Zealand plum blossom velvet antler powder,Jilin plum blossom deer velvet piece and Jilin horse deer velvet powder are selected as the monarch medicine to prepare the antler-kidney tonic decoction.The MTT proliferation test and cell scratch test showed that the three kinds of antler-kidney tonic decoction could inhibit the proliferation and migration of breast cancer cells and there was no significant difference.That is,the heavy metals and pesticide residues have no significant effect on the anti-breast cancer activity of the active ingredients of velvet antler.The effects of antler-kidney tonic decoction on the proliferation,migration and adhesion of MDA-MB-231 cells were detected by MTT,cell scratch and trypsin digestion.The expressions of Notch-1,Hes-1,Hey-1,Bcl-2 and Bax were detected by ELISA and Western Blot experiments.The results showed that the antler-kidney tonic decoction inhibited the proliferation and migration of MDA-MB-231 cells in vitro and inhibited the activity of Notch-1 signaling pathway.In addition,this study showed that the velvet antler polypeptide can inhibit the proliferation and migration of MDA-MB-231 and MCF-7 cells in breast cancer through similar experimental methods and also inhibit the activity of Notch-1 signaling pathway.It is presumed that the antler-kidney tonic decoction and velvet antler polypeptide may inhibit the proliferation and migration of breast cancer cells by regulating the Notch-1 signaling pathway,which may be a potential target and a new strategy for the treatment of breast cancer. |
