The deer velvet antler, a precious Chinese medicinal materials, refers tot the immature-ossification angle organization of the deer with thick feather hair. The velvet antler is a rich variety of natural factors of the cytokine library,which contains a variety of bioactive substances,they can health blood, strong bonesinvigorate kidney Yang they also can enhance immune function, promote organism metabolism and growth and other special effects. So the study on extraction process of pilose antler is of great theoretical significance and practical application.The research use alcalase alkaline protease、papain and trypsin separately which enzyme fresh deer antler to extract its effective components. The effect of enzymolysis was determined by determining the enzyme activity, degradation yield of fresh velvet antler,content of free amino acids and acids-soluble peptide in enzymatic hydrolysate. The results showed that the enzyme activity of alcalase alkaline protease was higher than other two enzymes,2×105U/g; degradation yield was highest,92.79%; The content of free amino acids was highest relatively is0.89mh/mL, the content of acids-soluble peptide was higher,53.8%; At the same time, the enzymatic solution is more clear, the color and liquidity is better respectively.In summary, alcalase alkaline protease was the optimal protease which was employed to enzymatic hydrolyze the free velvet antler. Fresh velvet antler was hydrolyzed by alcalase alkaline protease, the effect to degradation yield of fresh velvet antler by enzyme amount、substrate concentration、hydrolytic temperature、hydrolytic time and pH were determined by single factor experiment, screened important factors to determine the best technology condition by Orthogonal experiment: hydrolytic time6.0h, substrate concentration0.08g/mL, hydrolytic temperature65℃, pH9.0. And then the article studied on the ultrasonic-assisted enzymatic degradation processes of fresh velvet antler and liquid-solid ratio、enzyme amount ultrasonic power、ultrasonic temperature and ultrasonic time on degradation yield, determined the best technology condition of the technological process:liquid-solid ratio20, enzyme amount1.5%, ultrasonic power150W, ultrasonic temperature45℃, ultrasonic time40min, Under the condition, the degradation yield of Fresh velvet antler can reach to89.24%, Advantages of the method are that time is shorter and the extraction yield is higher.Lipids are obtained by using petroleum ether solvent extracted from enzymatic hydrolysates,detected and analyzed by chromatography-mass spectrometry, linoleic acid, linolenic acid, stearic acid etc10kinds of fatty acids were identified, the relative content of linolenic acid was high,41.232%. The molecular weight of Peptide in the layer was determined by ultrafiltration which mainly concentrated in the less than10000, when degradation yield is90%, the content of micromolecule peptide (M≤1000) can reach up to49.6%; separated by732strongly acidic cation exchange resin and studied on its separation condition, under optimal conditions, high-performance liquid chromatography was employed to detect and analyze the isolation products, arginine,lysine,leucine etc15kinds of amino acids were identified in enzymatic hydrolysate, of which, the content of arginine,lysine were the highest,138.20mg/100g,42.45m/100g respectively, they could be employed to manufacture high nutrient solution of nutritive value, make a great contribution to increase varieties of health food and good health.
