Chicken,duck meat and other poultry meat are rich in protein and other nutrition which are good to human body.And they have been become one of the daily food for people.However,many antibiotics remain in the animal body when raising animals.The antibiotic drugs remaining in poultry meat are likely to accumulate in human body.Therefore they are threat to people's health.Surface-enhanced Raman spectroscopy?SERS?and adaptive iterative re-weighted penalized least squares?air-PLS?were used in this research.Au nanoparticles and NaCl solution were used as SERS enhanced substrate for the rapid detection of AMOZ and AHD residues in poultry meat in the research.The main research contents are as follows:?1?The extraction process of poultry meat were optimized to shorten the experimental time and improve the experimental efficiency.Through the study on SERS of AMOZ standard solution,the Raman characteristic peaks of AMOZ were determined and their vibrational assignments were analyzed.Comparing with the characteristic peaks of poultry meat extract and poultry meat extract containing AMOZ,the SERS characteristic peak for the detection of AMOZ residues in poultry meat were determined.The optimum experimental conditions were determined by using the single factor experiment according to the characteristic peak intensity of AMOZ in chicken at 801 cm-1.On the basis of these conditions,the peak intensity ratio of AMOZ residues in chicken at 801 cm-1 and 1031cm-1 were quantitatively analyzed.There was a good linear relationship when the concentration range of AMOZ residues in chicken extract was 0.127 mg/L.The linear equation was y=0.1509x+0.4574,and the coefficient of determination?R2?was 0.9961.The minimum detection limit was 0.1 mg/L.Recovery rate of AMOZ residues in chicken extract was 82%122%.The optimum experimental conditions were determined according to the characteristic peak intensity of AMOZ in duck meat at 801 cm-1.On the basis,the linear model were established when the concentration range of AMOZ residues in duck meat and the peak intensity of duck meat extract containing AMOZ at 801 cm-1 or the peak intensity specific value at 801 and 1030.cm-1 or the peak area at 801 cm-1.And the recovery rate was tested.The model of SERS intensity at the characteristic peak 801 cm-1 was best.And the linear equation was y=224.99x+718.83,and the R2 was 0.9961.The minimum detection limit was 0.5 mg/L.And the recovery rate of AMOZ residues in duck meat extract was97%119%.?2?Through the study on SERS of AHD standard solution,the Raman characteristic peaks of AHD were determined and their vibrational assignments were analyzed.Comparing with the characteristic peaks of poultry meat extract and poultry meat extract containing AHD,the SERS characteristic peak of AHD residues detection in poultry meat at 606 and 1213 cm-11 were determined.The optimum experimental conditions were determined according to the characteristic peak intensity of AHD at 606 cm-1 in poultry meat.On the basis,the linear regression equation was established when the concentration range of AHD residues in poultry meat and the peak intensity of poultry meat extract containing AHD at 606 and 1213 cm-1.The linear equation of AHD residues in chicken at 606 cm-1 was y=263.19x+2042.6,and the R2 was 0.9955.The minimum detection limit was 2 mg/L.Recovery rate of AMOZ residues in chicken extract was 75%120%.The linear equation of AHD residues in duck meat at 606 cm-11 was y=528.56x+965.6,and the R2 was 0.9763.The minimum detection limit was 1 mg/L.And the recovery rate of AMOZ residues in duck meat extract was76%122%.But the linear equation of AHD residues in chicken at 1213 cm-1 was y=180.07x+591.33,and the R2 was 0.9908.The minimum detection limit was 2 mg/L.Recovery rate of AMOZ residues in chicken extract was 69%105%.The linear equation of AHD residues in duck meat at 1213 cm-11 was y=377.37x+462.98,and the R2 was0.9915.The minimum detection limit was 1 mg/L.And the recovery rate of AMOZ residues in duck meat extract was 79%116%.?3?A method was found to extract the AHD and AMOZ residues in poultry meat at the same time.The spectrum was collected by SERS technique.The background interference was eliminated by air-PLS and the data was preprocessed by standard normalization.The samples of four groups?sample of poultry meat extract containing AHD,sample of poultry meat extract containing AMOZ,sample of poultry meat extract containing AHD+AMOZ,sample of poultry meat extract?were marked in sequence and the model of cognition was established by combining the principal component analysis and linear discriminant analysis.In the model for the four groups samples of AHD and AMOZ residues in chicken,the correct discriminant rate of the calibration set was 90.48%and the correct discriminant rate of the prediction set was 94.29%,while in the model for the four groups samples of AHD and AMOZ residues in duck meat,the correct discriminant rate of the calibration set was 94.29%and the correct discriminant rate of the prediction set was95.71%.The research showed that the method was feasible to the rapid detection of AMOZ and AHD residues in poultry meat by SERS.And the samples of AHD and AMOZ residues in poultry meat could be identified effectively.
