In this paper,the microbial diversity is briefly introduced,and then two kinds of research methods of soil microbial diversity are described:biological or chemical based methods and molecular biology based methods.Then,high-throughput sequencing technology for microbial diversity was analyzed,and the application of high-throughput sequencing technology in microbial diversity was summarized.The soil microorganisms were treated with no-tillage film series,shallow layer fertilization series and straw-saving irrigation series from Inner Mongolia saline soil,respectively.The soil microorganisms were analysed by culturable microbial analysis or high-throughput sequencing.The culturable microbial experiment results showed that no-tillage and shallow tillage treatments could effectively increase the number and type of microorganisms in the saline soil,and 10%of conventional irrigation could be saved after the laying of straw.The results of high-throughput sequencing showed that the diversity of microbial diversity was higher in no-tillage film mulching treatment in saline-alkali soil,and the microbial diversity was lower in shallow layer.In addition,a bacterial strain NJ-Z5 T isolated from soil near the Antarctic Great Wall Station through the laboratory was systematically classified in this paper.The 16S rRNA gene sequence similarities of strain NJ-Z5T to the most closely related specie Arthrobacter sulfonivorans JCM13520T is 98.07%.The DNA-DNA relatedness values of strain NJ-Z5T with Arthrobacter sulfonivorans JCM13520T was 47.23%,which were below the recommended criterion of 70%for species delineation.Strain NJ-Z5T was Gram-positive,aerobic,non-mobility and rod-shaped,its colonies were ivory-white,it was found to grow at NaCl concentrations of 0-1%w/v,temperatures of 4-28??optimum 25??and pH6.0-8.0?optimum pH 7.0?.Positive for nitrate reduction,gelatinase,catalase,hydrolyze arginine,esculin,and urea.Negative for oxidase,H2S production,starch hydrolysis,Voges–Proskauer test and MR test.The predominant menaquinone was MK-9?H2?,MK-9,MK-10?H2?and MK-8?H2?,the major fatty acids were anteiso-C15:0?iso-C16:0?anteiso-C17:0?C16:1 w7c/16:1?6c and iso-C15:0,the polar lipid acids were DPG,PG,GL and UPL.Phylogenetic,chemotaxonomic and phenotypic analyses indicated that strain NJ-Z5T represents a novel species within the genus Arthrobacte,for which the name Arthrobacter albocanus sp.nov.is proposed.
