The rapid change of the natural environment will bring all kinds of stress,directly or indirectly affect the life of plants.Plants develop heritable and relatively stable adaptive mechanisms in the long run of adversity.As the responder protein of Ca2+,Calcium dependent protein kinases(CDPKs/CPKs)is widely found in plants and some protists.CPKs play a key role in signal transductions.Moreover,CPKs are essential in plant normal growth,resistance anddevelopment to adversity.Caragana intermedia is a good afforestation tree species to transform desertification areas of north China,which is tolerant to cold,drought,high temperature,salt and alkali.In this study,C.intermedia was used as a material to clone a CiCPK10 gene by some related technologies,and its primary functionunder different stress conditions were studied.The results are as follows:1.The cDNA of the CiCPK10 was 2763bp in length,and the open reading frame(ORF)was 1662bp long,encoding 553 amino acids.2.Under different stress conditions,Real-time fluorescence quantitative PCR was used to detect the expression of CiCPK10,and the results showed that it was induced by drought and ABA treatment,suggesting that it might have functional components that respond to the stress of adversity and corresponding signaling pathway.3.The overexpression vector p35s::CiCPK10-HA was constructed and was transformed into wild Arabidopsis thaliana.The expression of CiCPK10 in the homozygous lines of transgenic A.thaliana increased in various degrees.4.The water loss rate of the 2-week-old CiCPK10 over-expressed lines was lower than that of the control,and showed a significantly higher survival rate than the controlunder drought or 250 mM NaCl treatments.5.Under 100 ?M ABA or 250 mM NaCl treatments,the transcription level of stress-related genes was up-regulated in all over-expressed lines compared to the control.To sum up,CiCPK10 positively regulates the tolerance of the transgenic Arabidopsis to drought and salt stress.
