Lipids participate in the process of regulating the body water balance of insects,thereby maintaining their normal physiological activities,especially resisting the adverse environments such as high temperature and drought.Apolipoprotein D(Apo D)and Very low density lipoprotein(VLDLR)play an important role in the lipid metabolism and transportation of insects.This paper intends to study the gene expression and gene function of Tm Apo D and Tm VLDLR genes through gene cloning,q RT-PCR and RNAi,and to explore the relationship between these two genes and the body water balance of Tenebrio molitor.In the process,the ability to adapt to external stress such as humidity is laid the foundation.The experimental results obtained are as follows:The Tm Apo D and Tm VLDLR genes containing the full-length c DNA were amplified.The results of gene expression characteristics showed that: Tm Apo D and Tm VLDLR genes were highly expressed in fat body and head,respectively,which were significantly different from other tissue parts;while Tm Apo D and Tm VLDLR genes were highly expressed in female adult stage and the first instar larval stage,respectively,and were significantly different from other developmental stages.It indicated that the expression of Tm Apo D and Tm VLDLR genes of T.molitor was tissue-specific and development-stage specific.Low humidity stress caused the relative expression of Tm Apo D gene of T.molitor was significantly higher than that of the control group.Silencing the Tm Apo D gene using RNAi technology would lead to a very significant increase in the mortality of mature larvae.After RNAi treatment,feeding under low humidity stress would cause the mortality of the mature larvae to be significantly higher than that of the control group,indicated that the Tm Apo D gene was involved in the response process to low humidity stress.From the second day of RNAi treatment,the daily weight loss of the mature larvae was significantly higher than that of the control group;and the water loss rate of the experimental group was significantly elevated,indicated that the Tm Apo D gene will affect the growth and development and participate in regulating the water balance of the mealworm body.The relative expression of Tm VLDLR gene of T.molitor showed a dynamic change under low humidity stress,in which the poles on Day 1 and Day 5 decreased significantly,while the poles on Day 3 increased significantly.Silencing Tm VLDLR gene could cause the mortality of the mature larvae to be significantly increased,but low humidity stress had no significant effect on the mortality.In addition,RNAi treatment could cause the dailyweight loss of the mature larvae to be significantly higher than that of the control group;however,had no significant effect on the water loss rate.The above test results show that the Tm VLDLR gene could affect the growth and development of T.molitor,but not regulat the water balance.
