Larvae of Tenebrio molitor is rich in nutrients, for comprehensive utilization and to increase the added value, this paper from the lipid, protein and chitin three aspects of balance of extraction by mild Soxhlet extraction method to extract the oil, then Orthogonal test and response surface method of solvent extraction of protein and the acetic acid extraction of chitin test, draw the following conclusions:1. Under the premise of quality protection of Tenebrio molitor protein and chitin, grease the step 2 treatment method. Firstly, all of Tenebrio molitor pretreatment, determine water standard between 1-2%, followed by different temperature different ways to moderate the Soxhlet extraction method to extract oil and extraction rate of variance analysis and Kay method and Test method of determination of protein content in the degreasing before and after. Results:comprehensive consider the protection of protein and taking into account the grease is the optimum combination for the first time to water the best temperature to 65?, a second extraction optimum mode to 45?, the oil extraction 19.99% rate. The extraction rate was 16.12%, protein extraction rate was 3.68%, the test method of mass concentration 0.3032mg/mL(Bradform method).2. On the premise of protecting the quality of chitin under Tenebrio molitor (NaCl, NaOH, solvent, ethanol) extraction of Tenebrio molitor protein three, solvent results were compared, the best NaOH extraction of Tenebrio molitor protein effect.4 main factors influencing selection of 3 kinds of solvents of Tenebrio molitor protein single factor test of Orthogonal test, combined with Orthogonal design and Box-Behnken analysis the central composite design principle of fitting model, optimize the processing conditions, and consider the determination process of senior high school entrance examination method of BCA and protein content, the best solution is:(1) NaCl:1.0% concentration, temperature 40?, time 1h, solid-liquid ratio 1:14g/mL, extraction rate of protein was 49.37%, the concentration of 0.2112mg/mL (Bradform method) and 0.4953mg/mL (BCA method).(2) NaOH concentration is 2%, temperature 80?, time 1h, solid-liquid ratio 1:12g/mL, extraction rate of protein was 88.55%, the concentration of 0.2342mg/mL (Bradform method) and 1.1776mg/mL (BCA method). (3) ethanol:30% concentration, the temperature of 30?, time 2h, solid-liquid ratio 1:18g/mL, protein extraction rate is 40.44%, the mass concentration were 0.0854mg/mL (Bradform method) and 0.1796mg/mL (BCA method). In addition, two kinds of composite extraction method can obtain more proteins, respectively, for NaCl+NaOH protein extraction rate was 85.71%, the mass concentration were 0.201mg/mL (Bradform method) and 0.6227mg/mL (BCA method) and NaCl+ ethanol protein extraction rate 54.51% and the concentration were 0.1083mg/mL (Bradform method) and 0.2258mg/mL (BCA method).3. Solvent used in combination with acetic acid extraction of Tenebrio molitor chitin: chitin extraction rate respectively is NaCl:17.24%, NaOH insect powder:14.62, NaOH residue:15.43% ethanol:16.23%, NaCl+NaOH:23.15% and NaCl+ethanol:34.88%.6 method residual ash less than 1%, and ensure chitin quality to food grade requirements.
