The frequent occurrence of respiratory diseases is closely related with the change of respiratory tract floar.The theory that respiratory diseases are regulated and controled by respiratory flora is increasingly recognized by people.It is extremely important to clarify the mechanism of the disease occurrence,development and transformation by exploring the microecological distribution of respiratory tract.At the same time,it provides valuable reference for the rational use of antibiotics and vaccine prevention,and also provides new ideas for the effective treatment of respiratory diseases.In this study,Karakul Sheep in Aksu area were selected as the research object,and the microbial fine and fungal community structure of sheep respiratory system was studied by genome sequencing technology.In the experiment,the lung samples of sheep with obvious pathological changes were collected and compared with those of healthy sheep;the complete respiratory system was divided into four parts: the nasal cavity,the upper end of the trachea,the lower end of the trachea and the lung,and the samples were collected separately;the nasal swab samples were taken from the nasal cavity,and the mucus samples from the respiratory tract were taken from other parts to detect the distribution of fine and fungal groups in the corresponding parts.The results showed that in the 38 samples there were 2 Kingdom,27 phylum,44 class,98 orders,178 families and 462 genus in bacteria level,and 1 Kingdom,16 phylum,47 classes,114 orders,264 families and 494 genus in fungi level.The dominant bacteria species in respiratory system of healthy sheeps were Acinetobacter.lwoffii,Psychrobacter.faecalis,Lactobacillus.iners,Moraxella.ovis,Acinetobacter.haemolyticus,Alicycliphilus.denitrificans.The dominant bacteria species in the respiratory tract of sick sheeps were Helicobacter.SP.CLO-3,Mycoplasma.ovipneumoniae,Mannheimia haemolytica,Moraxella bovoculi,Psychrobacter faecalis,Isobacteria.The dominant fungi in respiratory tract of healthy sheep were as follows: Issatchenkia.orientalis,Fusarium oxysporum,Mortierella alpina,Lasiodiplodia theobromae,Alternaria alternata,Candida albicans,Isobacteria.Further analysis showed that compared with the healthy sheep,the number of Helicobacter,Mycoplasma ovipneumoniae,haemolyticus mansoni,Moraxella,psychrophilic,Pseudomonas aeruginosa,Prevotella,Ureaplasma,Streptococcus and rumen leivotella were significantly increased in the sheep with respiratory diseases;these bacteria groups were basically maintained at 0.1-1.8% in the respiratory system of healthy sheep;the number of bacteria in the sheep with respiratory diseases was significantly increased After the disease,the amount of some pathogenic bacteria can reach between 2-10%,and even some pathogenic bacteria can reach as high as 60-70%.From the results of lefse analysis,we can see that the three pathogenic microorganisms such morcelli,mycoplasma and Streptococcus have the greatest impact,and may play a major regulatory role in the process of disease occurrence and development.Through the study,we aim to figure out the pathogeny meachism of the microecology to treat the respiratory diseases;at the same time,we can provide some reference for the abuse of antibiotics,guidance of medication and other aspects,and provide some new ideas for effectively reducing the incidence of respiratory diseases.
