Galleria mellonella belongs Lepidoptera,Pyralidae,Galleriinae and Galleria,Widely distributed around the world,it is an important pest of the beekeeping industry.In the early days,as an important pest of the beekeeping industry,it was mainly the study of its prevention and treatment methods.In recent years,with the deepening of research,people have realized that the wax is rich in protein,short in growth cycle,rich in food sources,easy to breed and edible,and more importantly in medicine,pharmacy,physiology,agronomy.Disciplinary fields play an irreplaceable role.At present,a large number of research results have been obtained on the research of larvae,parasitic wasps,new cryptococcal bacteria,antibacterial peptides,and antibacterial immune mechanisms.Recently,bacterial strains isolated from the larval gut of the researchers have confirmed that they can feed and decompose polyethylene plastics in a laboratory environment,which has made the wax larvae's function of degrading plastics widely concerned.This study utilizes traditional culturable techniques combined with ITS rDNA and 16 S rRNA gene sequence analysis techniques to treat various parts of the Galleria in three different feeding conditions(plasticized-fed,feed-fed,wild)The culturable microorganisms(fungi,bacteria)of the tract,mouth,feces,body surface and silk are identified by molecular biology,and the microbial community structure and its diversity are evaluated.On the one hand,this research further supplements and enriches the research content of the G.mellonella,laying a certain theoretical foundation for the biological research related to the G.mellonella;on the other hand,it is not only beneficial to the development and utilization of insect resources,but also beneficial to insects.This special "habitat" mines microbial resources with special functions,which are used to open up new ways of biotransformation and develop new biomass energy.In this study,72 strains of fungi were isolated from different parts of the three different feeding conditions,belonging to 2 Phylum,Ascomycota and Basidiomycota.The ascomycete was the dominant,and 58 fungi were isolated.,accounting for 81.69%.There are 7 classes,12 orders and 21 genera.The dominant one is Penicillium,accounting for 19.72%,followed by Aspergillus,accounting for 15.49%.For the isolation rates of different parts of the different feeding conditions,the diversity index(Shannon-Wiener index,Simpson index Margalef index,Pielou index)analysis led to the following three main conclusions.1)The isolation of culturable microbial strains is quite different due to different feeding conditions and separation sites: for the overall analysis show that the separation rate of bacteria(173 strains)is significant for three different feeding conditions.More than the isolation rate of fungi(71 strains).For the different parts of the analysis show that the cultured fungus is more easily obtained from the body surface,while the bacteria are obtained from the intestinal site.2)The composition and distribution of the community have certain universality and particularity: the fungi of Aspergillus and Penicillium are common in the feces,body surface and silk of the G.mellonella,the bacteria Enterococcus,Bacillus is ubiquitous in various parts,and all are dominant genus;while the fungi Cladosporium is only distributed in the mouthparts,and the bacteria Lysinibacillus,Psychrobacillus are only distributed in the silk,and The separation rate is low.3)Analysis of community diversity showed that the highest diversity of fungal community was feed-fed G.mellonella,and the highest diversity of bacterial community was plastic domestication;and for isolated parts,the body surface of fungal diversity was highest.The bacterial diversity is highest in the intestines.
