Purpose:Detection of long non-coding RNA(lncRNA)expression profiles in autologous arteriovenous fistula(AVF)outflow tract stenosis in patients with renal failure by high-throughput sequencing,and to explore lncRNA and AVF outflow tract stenosis relationship.Methods:From February to June 2017,the Department of Nephrology,the First Affiliated Hospital of Nanchang University,diagnosed 28 cases of vascular tissue in the reoperation of AVF patients with disabilities(experimental group).The inclusion criteria were guinea for 3 months;Flow rate ? 200 ml/min;internal stenosis degree greater than 50%,exclude obvious thrombosis;upper limb vascular ultrasound examination data intact.The control group was selected from 75 patients with vasospasm who had the first action of intravenous sputum in the same institution.The inclusion criteria were those with an outflow vascular diameter of ?1.6 mm and no vascular stenosis or other vascular diseases.Total RNA was extracted from 103 samples by Trizol reagent,and 4 cases in the experimental group and the control group were selected respectively.The differences in lncRNA and mRNA expression profiles were detected by high-throughput sequencing,and then differentially expressed lncRNA and mRNA were screened.Cluster analysis,GO analysis,KEGG pathway analysis;the RNA-seq results were verified by real-time RT-PCR(qRT-PCR);The amount of LINC01615 was highly expressed in the arteriovenous fistula outflow tract stenosis.Results:Compared with the control group,lncRNA or mRNA with a change of expression?2 fold and a statistically significant difference(P < 0.05)was considered to be a differentially expressed lncRNA or mRNA.(1)There were 247 lncRNA expression changes?2 times,141 in the experimental group were up-regulated?2times,and 106 were down-regulated?2 times;(2)There were 1386 mRNA expression changes?2 times,874 in the experimental group were up-regulated?2 times,and 512 were down-regulated?2 times;(3)GO analysis suggested that significantly different expression of lncRNA is mainly involved in cell differentiation and apoptosis;angiogenesis,vascular development,vascular system development,etc;(4)KEGG pathway analysis suggested that the differentially expressed genes were most commented on the PI3K-AKT signaling pathway,and the most significant one was the TGF-? signaling pathway;(5)In the significantly differentially expressed ncRNA,qRT-PCR was used to verify the critical 9 lncRNA and 7 mRNA expression levels,and the results were consistent with RNA-seq;(6)Compared with the control group,LINC01615 was high expression in autologous arteriovenous fistula(AVF)outflow tract stenosis.Conclusion:There are significant differential expression of lncRNA in AVF in uremic hemodialysis patients.These lncRNA and some signaling pathways may play an important role in the occurrence and development of AVF outflow tract stenosis in uremic patients.This study may lay the foundation for the elucidation of the pathogenesis of AVF outflow tract stenosis and provide potential therapeutic targets for patients with AVF failure.
