There are a large number of non-coding RNA(nc RNA) sequences presented in the human genome sequence, the long non-coding RNA are a class of non-coding RNA, which are larger than 200 nt. Lnc RNAs are closely related to many human being’s physiological processes, especially play very important roles in the occurrence and prognosis process of many diseases. Due to the strong time- and tissue- specificity of lnc RNA expression, there is a great potential of novel lnc RNA prediction. As millions of transcripts are generated by next-generation sequencing, larger amount of lnc RNA have be identified, most of which have not been functionally characterized. How to associate a lnc RNA with its target m RNA, and how to indicate its potential function, are also our concern. lnc RNA-Seq is an effective technology that utilizes the capabilities of next-generation sequencing to study lnc RNA, while reference annotation-based assembly aids in the detection of novel isoforms, which has been applied in fundamental scientific research, clinical diagnosis, pharmacogenomics research and drug research and development, and so on.In many studies, the lncRNA-Seq has been used and achieved a lot of data and experimental conclusion. However, there are few reports on studies of lnc RNAs in response to ionizing radiation(IR).Basing on our the hypothesis that some lnc RNAs can be induced by IR, total RNAs of human cells are isolated after exposure to X-rays and heavy ion irradiation generated by the heavy ion research facility in Lanzhou and the cooling storage ring(HIRFL-CSR), and sequenced by lnc RNA-Seq technology. Through bioinformatics analysis, classification and annotation, we obtained manylnc RNAs which play a role in the IR-induced cellar responses. After exposed to different doses of irradiation, the expression levels of several special lnc RNAs are measured with q RT-PCR. To further confirmed the condition of lnc RNA response to IR,the expression levels of lnc RNAs in response to different kinds ionizing radiation in different cell lines were also tested by q RT-PCR. We obtained many IR-induced lnc RNA, and found out the key lnc RNAs responding to IR. We identified 647 kinds of known lnc RNAs and 381 kinds of novel non-coding sequences responding to ionizing radiation.This is the first report of lnc RNAs in response to IR and our findings put forward a new research direction in radiation biology.
