The intestine is the largest immune organ in the human body.The normal flora in the intestine is an important factor in maintaining intestinal homeostasis.They can regulate the body's lipid metabolism by secreting beneficial metabolites.When the flora is unbalanced,it will cause various diseases.Oral microecological preparations are an important means and a way to solve the imbalance of intestinal flora today.This study involves twice a week for 8 weeks in total of fecal collection and sequencing using High-throughput sequencing technology called Ion Torrent PGMTMfrom disease populations with constipation,diarrhea and comprehensive intestinal patients?including abnormal stools,bloating fecal abnormalities,insomnia,irregular bowel movements and irritable bowel syndrome?.It is worth mentioning that the populations took the Stachyose tetrahydrate?Sta?,Probiotics Power?PP?and Probotic Preparations?PPrs?for 8 weeks respectively.And bioinformatics and multivariate statistical methods were applied to deeply explore the changes in unbalanced intestinal flora structure.Supplemented by the test results of short-chain fatty acids,cholesterol and bile acid in feces,it was confirmed that these three types of microecological preparations were related to the regulation of intestinal flora and lipid metabolism,and identified key genus related to lipid metabolism.Finally,the correlation and information between the regulation of microecological preparations and the diversity of intestinal flora related to lipid metabolism was explained.Firstly,the sequencing of the fecal samples of 27 subjects showed that there were significant differences in gut microbiota diversity among healthy,constipation,diarrhea and comprehensive intestinal patients?p<0.05?,and the gut microbiota diversity of the three groups of patients was lower than that of the healthy group.Among them,the diversity of constipation populations was relatively high,and the diversity of intestinal flora in diarrhea populations was significantly lower?p<0.05?.At the level of the phylum,the intestinal flora of the four groups of people was still dominated by Bacteroidetes,Firmicutes,Proteobacteria and Actinobacteria,but the abundance of different individuals was different:Actinobacteria and Firmicutes in the constipation populations was higher,and Proteobacteria in the comprehensive intestinal patients was higher.The Firmicutes was higher in diarrhea populations,but the Bacteroidetes was lower.At the level of the family,in addition to the Alcaligenaceae in three disease groups were lower than the healthy group,the abundances of Laschnospiraceae,Clostridiaceae and Enterobacteriaceae were lower,Veillonellaceae was higher in the intestine of constipation,and the other two groups were opposite.Prevotellaceae,Porphyromonadaceae and Ruminococcaceae had lower abundance,Bacteroidaceae was higher in the intestine of diarrhea,and the other two groups were opposite.At the genus level,Bacteroides were more abundant,Ruminococcus,Parabacteroides,Oscillospira,Clostridium,Dialister and Prevotella were lower in diarrhea populations.Lachnospira and Coprococcus were less abundant,Veillonella and Megasphaera were higher in constipation populations.The Faecalibacterium was lower,the Mitsuokella and Citrobacter were higher in comprehensive intestinal patients.Moreover,Coprobacillus,Enterococcus and Parabacteroides may be the key genus causing constipation while Dialister,Paraprevotella and Veillonella may be the key genus causing diarrhea.Then,through the sequencing of fecal microflora under the intervention of microecological preparations,the detection of lipid metabolism indicators and the quantitative analysis of key bacterial populations,it was found that the effects of the three microecological preparations on the structure and diversity of intestinal flora were PPrs>PP>Sta.In addition,Sta could reduce the abundance of Bacteroides in diarrhea populations,increase the abundance of Prevotella and Faecalibacterium,among which Bacteroides was negatively correlated with bile acid,positively correlated with cholesterol,Prevotella was negatively correlated with acetic acid.Moreover,the content of Bifidobacterium spp,Lactobacillus spp and Faecalibacteriurn prausnitzi had been increasing,and they were continuously approaching the healthy level.PP had a role in adjusting to healthy abundance levels in Bacteroides,Sutterella,Faecalibacterium,Clostridium,Klebsiella,Megamonas and Dialister in populations with a comprehensive intestinal disease,but with a shorter duration of action.Among them,Bacteroides was negatively correlated with bile acid,cholesterol,acetic acid and propionic acid,while Klebsiella was positively correlated with them.Sutterella was negatively correlated with bile acid and propionic acid,positively correlated with cholesterol and acetic acid.Faecalibacterium was negatively correlated with bile acids,acetic acid and propionic acid,positively correlated with cholesterol,Blautia was positively correlated with acetic acid and propionic acid,and Megamonas was negatively correlated with acetic acid and positively with propionic acid.PPrs could reduce Bacteroides in constipation populations,raise Prevotella,and adjusted Dialister and Sutterella to healthy abundance levels.Among them,Sutterella was positively associated with bile acids,acetic acid,propionic acid,butyric acid,and negatively correlated with cholesterol.Prevotella was negatively correlated with acetic acid,propionic acid and butyric acid;and the amounts of Lactobacillus spp,Bacteroides,Bifidobacterium spp and Faecalibacteriurn prausnitzi continued to increase.And the amount of Bacteroides,Lactobacillus spp and Faecalibacteriurn prausnitzi were close to healthy levels after the intervention.In summary,there are significant differences in the bacterial structure and diversity of people with intestinal diseases,and microecological preparations can actively act on the intestinal tract and regulate various lipid metabolism indicators,and there is a correlation between the abundance of the flora and the content of various lipid metabolism indicators.The regulation effect of PPrs is better overall.Through the adjustment of intestinal micro-ecological diversity,the clinical symptoms of constipation,diarrhea and comprehensive disease have been better improved,and the regulation effect is obvious;Sta has the best effect on the regulation of diarrhea populations,PPrs has the best effect on constipation populations,and PP has a weak effect;Meanwhile,the study reveals that the three types of microecological preparations can regulate the abundance of intestinal flora related to lipid metabolism and regulate lipid metabolism.This indicates that the microecological preparation not only has the effect of significantly regulating the diversity of intestinal flora,ameliorating intestinal diseases,but also plays a role in regulating lipid metabolism related indicators.The study also reveals the commonality of the three types of microecological preparations:the effects are time-sensitive,and the effects cannot be sustained after the intervention is stopped,so that the patients are dependent on them.
