Amphibian frogs are an important indicator of ecological environment,among which Ranidae is an important species.The gut is the body's largest reservoir of bacteria and immune organs.In the process of host growth and development,many physiological and immune functions are improved with the assistance of intestinal flora.Studies have shown that the presence of normal bacteria in animals can stimulate the body's healthy immune system to maintain its own immunity.When intestinal bacteria are absent,it can cause the intestinal immune system to underdevelop.Amphibians are more sensitive to environmental changes than other groups due to their unique life history and high skin permeability.As a result,some scientists speculate that the environment has a significant influence on it.So how are the different Ranidae hosts related to the microbial community and the environment?In this study,Illumina high-throughput high-throughput sequencing technology was used to detect the structural characteristics of the intestinal flora of Rana chensinensis in yichun,Rana amurensis in yichun and Rana amurensis in Harbin.The T cells in the intestinal tissue sections were observed microscopically.meanwhile,the intestinal lysozyme activity was detected.In order to provide basic data for species protection and breeding of Ranidae,the structural characteristics and intestinal immune function differences of intestinal flora of different species of Ranidae and the same geographical population of Ranidae were compared.The sequencing results showed that Rana amurensis in Harbin and yichun and Rana chensinensis and Rana amurensis in yichun were highly similar to each other in intestinal microbial community structure.Rana chensinensis had higher diversity than Rana amurensis,and the predicted functional abundance of microbial gene Rana amurensis was generally higher than Rana chensinensis.Although the dominant bacteria with important functions in Ranidae are consistent,the specific proportion of the dominant bacteria in each sample is significantly different at different classification levels.ANAE staining and lysozyme test results showed that the number of T cells and the content of lysozyme,Rana chensinensis in yichun area<Rana amurensis in yichun area<Rana amurensis in Harbin area.To sum up,the difference in intestinal flora structure of amphibian frogs should be attributed to the combined effect of species and environmental factors,and it is speculated that Rana amurensis is more widely distributed than Rana chensinensis and has a relatively larger population,which may be related to the higher antimicrobial capacity of Rana amurensis and the more active microbial gene function.
