This study is the first systematic study of the Tanypodinae of Chironomidae in Wuling Mountain area.Wuling Mountain area is a hot area of biodiversity research in China and even in the world,but the study of Tanypodinae in this area is relatively weak.By consulting the literature,sampling in the field,combining the traditional morphology with modern molecular method,the diversity of Tanypodinae in this area were preliminarily studied.In this study,22 species belonging to 12 genera of Tanypodinae were summarized(Including this study),and the old records were identified and described again.Three new recorded genera in the region: Denopelopia,Zavrelimyia,Meropelopia.Seven new recorded species in Wuling Mountain area:Meropelopia sp.1,Denopelopia sp.1,Zavrelimyia sp.1,Ablabesmyia longistyla,Tanypus nakazatoi.Conchapelopia sp.1?Procladius sp.1?Besides,14 mitochondrial cytochrome coxidase subunit 1(COI)genes were obtained from the subfamilies of Tanypodinae,Two COI genes of 2 genera of Chironominae subfamily(Chironominae subfamily as outgroup),in order to be more effective in molecular identification,72 sequences of COI gene from NCBI database were downloaded and analyzed by bioinformatics.Based on the sequence of COI gene,the molecular species were obtained by the neighbor-joining tree,and the successful matching rate with morphological species was high.At the same time,the intraspecies and interspecific genetic distances of some subgroups of Tanypodinae were analyzed.The results of molecular analysis showed that the intraspecific genetic distance and the interspecific genetic distance were 0-0.040 and 0.001-0.134,respectively,indicating that the intraspecific genetic distance of some species was larger than that of the interspecific genetic distance,and there might be cryptic species in this genus,or there are homonyms and other phenomena between species,which need to be further studied.In this study,we explored the diversity of Tanypodinae subfamilies in Wuling Mountain Area,and verified the role of DNA barcodes in species identification.The results of this study filled the blank in the systematic study of the subfamilies of Tanypodinae in this area,and the molecular data expanded the barcode database of the subfamilies of Tanypodinae,which provided some basic data for the phylogenetic analysis of the subfamilies.
