In the process of cultured American ginseng in the farmland, the soil physical and chemical properties are gradually deteriorated. And soil nutrients can supply to plant adequately. It will cause many serious diseases. And the production of American ginseng is lower and the quality is poorer. In order to solve these problems and improve the soil fertilizer content of American ginseng cultivation soil, this paper designed six treatments with different bio-organic fertilizers. We add same amount of bean cake and corn stover to every treatment. This paper analyzed the soil nutrient, soil enzyme activity and soil microbial community composition with different treatments, combined with four-year-old American ginseng's yield and saponin content. The results showed that;(1)In the six bio-organic fertilizer soil improvement treatments of this experiment, treatment 3(0.45 kg·m-2 bean cake,2 kg·M-2 com stover,0.16 kg·m-2C,0.18 kg·m-2D,0.16 kg·m-2E)has the highest yield, reaching 15435.8 kg·hm-2.The trearment 3's American ginseng's total saponin content is highest. And Rb1 content is 2.738%,the Rb2 content is 0.139%,the Rb3 content is 0.065%,and the Re content is 0.198%.(2)With the growth period of American ginseng, the soil was gradually acidified. When American ginseng grew to the fourth year, the total nitrogen content of the six treated soils decreased by about 0.1 g kg-1,the total phosphorus content did not change significantly, and the total potassium content decreased 0.2-0.5 g kg-1.(3)Different treatments had little effect on soil catalase activity.Treatment 5 had significant effects on soil urease and phosphatase activity, and the six treatments had slightly increased soil invertase activity.(4)According to DGGE and high-through put sequencing technique, the common bacteria in all soils samples are:Chelatococcus.and Phaeospirillum.,Sphingomonas.and Sphingobium. In bacteria microbial community,Proteobacteria is the dominant bacteria. In the fungal microbial community,Ascomycota is the dominant fungi.Treatments 3 and 4 were more similar in both bacterial and fungal community structures.
