Objective: This research is aiming to establish Fujian Han population Power Plex 21 fluorescent multiplex amplification system 20 autosomal STR loci on the DNA database,to understand the possibility of genotype ratio between 20 STR loci in two unrelated individuals in the DNA database,and a discussion of core STR loci in Fujian Han population DNA database is included.Methods: ? Collect peripheral blood 3u L and extract DNA from total of 5650 unrelated individuals samples in our laboratory for DNA Parentage Testing on the Han nationality in Fujian.? Research on gentyping of 20 autosomal STR loci and 1 sex loci within 5650 unrelated individuals by using PP21 detection system,which is based on PCR composite amplification and capillary electrophoresis..? Input the above 5650 unrelated individuals of genotype results and sample informationinto PP21 detection system.Establish the DNA database in Fujian Han population of 20 autosomal STR loci.? The 5650 independent individuals samples were compared to the independent genotype of the DNA database by using the above comparison system to work out the possibility of the complete ratio of the 20 autosomal STR loci Calculate Combined paternity index of the duosbase on assumption that if duos CPI is greater than 10000,kinship supporting;if duos CPI is less than 0.0001,no kinship;and if duos CPI is between 0.0001 and 10000,possibility for both..? Regarding the duos of CPI > 0.0001,additional parameter of Investigator HD plex fluorescent multiplex amplification system is added for further test.Calculate combined paternity index of the 30 autosomal STR loci,same resulthas been achived.? There is chance that the duos of 30 autosomal STR loci is unable to eliminate the possibility genetic kinship,addtional parameter Investigator Arugs X-12 multiplex amplification system is considered for further tests.In combination with the age statistical analysis,further confirmation on unrelated individuals can be reached.Results: ? The establishment of Fujian tracking system in Han population of 5650 unrelated individuals samples of 20 autosomal STR loci in DNA database.? Results of testing STR loci on 5650 unrelated individuals :The genotype comparison that 20 autosomal STR loci of 5650 unrelated individuals was 23,the matching ratio of 20 STR loci was 0.41%(23/5650).Furthermore,CPI of 20 STR loci in these 23 individuals was calculated,8 cases show genetic kinship cannot be eliminated;15 cases support genetic kinship.? 10 extra autosomal STR loci by HD detection system testing(A total of 30 autosomal STR loci)applied to the above 23 samples,2 cases supported the genetic kinship,in which,1 were presumed to have 30 autosomal STR loci identicaland the other 1 had 1 mismatched loci.The remaining 21 cases concluded no evidence of generic existence.The matching ratio was 0.02%(1/5650).? With combination of X-12 detecting system and the age statistical analysis,the above 2 cases which can not be eliminated by 30 autosomal STR loci,resulted no evidence of genetic kinship.The matching ratio was zero.Conclusion: ? The 20 autosomal STR loci in the PP21 detection system are the core STR loci and are suitablefor DNA database of the Han population in Fujian.? In Fujian Han population DNA database,two unrelated individuals tested based on 20 autosomal STR loci were in accordance with Mendel's genetic law,the matching ratio is 0.41%.When testing autosomal STR loci increased to 30,the numberdropped to 0.02%,and further dropped to zero when 12 X-STR loci is included in test.? 30 autosomal STR loci detection is recommended for duos case especially in tracking missing persons.For those cases that still cannot be eliminated,the sex chromosome STR loci should be further detected to auxiliary result judgment.
