Skin aging is the most obvious external manifestation of aging.Usually,the entire skin gradually begins to show signs of dryness,wrinkles,uneven pigmentation,senile plaques,freckles or liver spots(sun spots),and delayed wound healing.Skin aging is a natural process that is caused by both endogenous changes and exogenous damage.The appearance of skin aging is mainly due to UV-induced ROS response in skin cells,activation of downstream pathways,which affect the expression of collagen I(Col-1)and matrix metalloproteinase I(MMP-1).The loss of Col-1 caused wrinkles and other aging characteristics.Human epidermal growth factor(hEGF)is an important active ingredient currently used in clinical skin repair,regeneration and anti-aging.However,the low transdermal bioavailability and high production cost due to the large molecular weight and complex structure greatly limit its medical and commercial applications.This paper aims to design a kind of hEGF-like functional short peptides,which can improve the anti-aging repair activity at the same time,make it as small as possible,easy to absorb,easy to synthesize,and reduce costs.Therefore,using the four anti-oxidation peptide libraries established earlier in the laboratory as a reference template,a total of nine transformants were obtained through molecular design and optimization.Subsequently,a series of biological models were screened at nine in vitro and in vivo levels,respectively,including the detection of antioxidant capacity,cytotoxicity assays,detection of the transcriptional activation of Col-1,and validation of animal skin repair models.It has been consistently demonstrated that P-WH is the best modified body that combines strong antioxidant capacity,low toxicity,and promotes collagen transcription and skin repair.Through two animal fold models,it was proved that 100 ?g/m L P-WH can not only inhibit the formation of wrinkles,but also have the effect of repairing and reversing the already existing folds,and have been confirmed by the skin elasticity test and skin thickness measurement experiment.Further research on the skin repair mechanism of P-WH revealed that P-WH can significantly reduce ROS content in human skin fibroblasts,up-regulate the expression of type I collagen Col-1,and decrease the expression of its hydrolase MMP-1.Involves multiple molecular mechanisms;Finally,P-WH was evaluated for safety and stability,demonstrating that P-WH has excellent development prospects with low toxicity,low hemolysis,no sensitization activity,and good stability.
