Hainan eld's deer is a national class I protected species.Gut microorganisms are important for its survival.The microbial community existing in the intestine is the result of long-term evolution of the species,and is closely related to animal food,body immunity and nutritional needs.The probiotic bacillus was screened by antagonistic experiment,and high throughput sequencing was used to analyze the intestinal micro flora of Hainan eld's deer by 16S rRNA sequencing,and the dominant bacteria in intestinal flora were excavated.The results were as follows:1.Probiotic bacillus were isolated from the feces of the healthy Hainan eld's deers.The screening strains were identified by the comprehensive morphology,physiological and biochemical analysis and molecular identification.Finally,the Bacillus probiotic 2-1-1 was identified as Bacillus spp.and Y17 was Bacillus pumilus.2.Through the construction of the 16S rRNA gene clone library and the phylogenetic analysis,the structure and diversity of 16 samples collected from the healthy adult Hainan eld's deer in Guangzhou zoo were analyzed.The results showed that the gut microorganism in Hainan deer was divided into 18 microbes,phylum,and the Firmicutes and Bacteroidetes were the core bacterias,accounting for 50.49%and 36.52%respectively.The top 10 genera of the Hainan eld's deer were 5-7N15,7.181%;Prevotella,3.159%;Treponema,2.788%;Succinivibrio,2.545%;Oscillospira,2.304%,CF231,1.843%;Ruminococcus,1.652%;Akkermansia,1.439%;Methanobrevibacter,1.340%and Clostridium,1.333%.3.By comparing the gut microbiome of Hainan eld's deer with different sex,17 species with significant difference(p_value<0.05)were found by T-test test.In group A,the genus with higher abundance were:Desulfovibrio,Ruminobacter,Clostridium,p-75-a5,Oxalobacter,and 02D06,Escherichia,YRC22,BF311,SHD-231,and Lachnospira,and higher abundance in the group B were:5-7N15,Butyricicoccus,Phascolarctobacterium,Anaeroplasma,Dorea,Blautia.
