Nuprl is a small nuclear protein with diverse physiological and biological functions.Nuprl plays an important role in stress response and cancer,and which is transcriptionally activated by TGF? via the SMAD signaling pathway.Nuprl regulates the physiological activities of cells through the TGF? signaling pathway and involved in the regulation of endoplasmic reticulum stress pathways,which plays an important role in apoptosis.To study the expression of Nuprl mRNA in mouse uterus during early pregnancy,the method of in situ hybridization was used to investigate Nuprl mRNA expression in animal models that include early pregnancy,pseudopregnancy,delayed implantation and activation,artificial decidualization and hormonal treatments.The relative expression levels of Nuprl mRNA were detected in early pregnancy and pseudopregnancy using real-time PCR.During mouse early pregnancy,the signal of Nuprl mRNA was detected in luminal epithelium and glandular epithelium during the days of 1?4 and in the decidua area during the days of 5?8.The signal was detected in luminal epithelium and glandular epithelium of the mouse uterus in the delayed implantation,which is similar to the results of early pregnancy on day 4.The signal was detected in decidua in the model of activation,which is similar to the results of early pregnancy on day 5.The expression of Nuprl mRNA in the model of artificial decidualization was detected in decidua area.In the control of artificial decidualization the slight signal appeared in luminal epithelium and glandular epithelium of the mouse uterus.After treated with oestrogen the signal appeared in luminal epithelium and glandular epithelium of the mouse uterus,and the signal was enhanced.After treated with progesterone the signal appeared in luminal epithelium and glandular epithelium of the mouse uterus.After treated with both of oestrogen and progesterone the expression of the signal was not changed significantly.Real-time PCR result showed that the relative expression of day 2 is higher than other days in early pregnancy,the level of Nuprl mRNA decreased from day 2 to 3,increased from day 3 to 4,remained close from day 5 to 8.The relative expression of day 2 is higher than other days in pseudopregnancy.The results indicated that the expression of Nuprl mRNA in mouse uterus is related to the process of mouse early pregnancy.The expression of signal in luminal epithelium and glandular epithelium of the mouse uterus may be regulated by hormones.Nuprl mRNA expression in uterine stroma is associated with decidualization and active blastocysts.
