China giant salamander(Andrias davidianus)are two key protected rare animal in China,it is a transitional amphibian from aquatic to terrestrial,it is called "living fossil",and is a valuable model animal in scientific research.The skin of the giant salamander is the first effective barrier between the body and the environment,is the first defense system of the body,and plays an important role in the process of regulating life activities.In addition,the giant salamander has a strong hunger tolerance,in the absence of food,the giant salamander can survive several months or even several years.The study on the hunger tolerance of giant salamander,and uncover the mystery of its fasting endurance,will greatly promote the development of technology and level of human medicine and other aspects.This experiment used the Chinese giant salamander skin as material,based on experimental techniques such as isotope labeling,mass spectrometry,two-dimensional electrophoresis etc,combined with IPA,iTRAQ and other analytical methods,from the perspective of proteomics,we studied the abundance of protein related to stress response,inflammatory reaction and immune reaction physiological activity,the ratio value of expression abundance and the change of physiological activity.(1)The expression of proteins participated in immune-related biological process in Chinese giant salamander:The proteins participated in immune-related biological process in Chinese giant salamander were identified.The biological process what the proteins mainly involved in include the cell recruitment related to innate immune inflammatory response,removal of antibody-antigen complex and antigen labeling,antigen presentation and processing of humoral immunity,Complement activation and B cell activation and proliferation,the activation and proliferation of T cells of cellular immunity,the activation of complement and other physiological activities related to cellular immunity.(2)The expression of proteins participated in immune-related signaling pathways in Chinese giant salamander:The proteins participated in immune-related signaling pathway in Chinese giant salamander were identified.The identified proteins involved in 31 signaling pathways include PLC,Paxillin,JAK-STAT,GPCR,NRF2,HER-2,IL-12,IL-22,IL-10,TNFR1/2,Reelin,CRH,PI3K/Akt,HIF,IL-6,ERK5,HMGB1,IL-2,IL-3,IL-8,Erb,FLT3,NF-?B,PKC?,TREM1,IL-1,IL-4,Myc Mediated Apoptosis,Trk,OSM and TLR.(3)Effect of fasting on the expression of immune response-related proteins in Chinese giant salamander:this study through the detection of Chinese giant salamander fasted for 0,1,2,3,4,5,7,8,9,10,11 M etc total of 11 time points expression of immune response activities related protein abundance of species found that,167 proteins are directly involved in the immune response,involved in the physiological activities related to the immunoreaction of recruitment,antigen labeling,and the clearance of antibody antigen complexes related to inflammatory response related cells.209 proteins involved in regulating the immune response signaling pathway,including 31 signal pathways associated with immune response such as cCRH(corticotropin releasing hormone),kappa BNF-kappa B(nuclear factor),BPI3K/Akt(phosphatidyl inositol 3 kinase / protein kinase).Protein is the embodiment of life activity.To study the role of skin in hunger tolerance from the proteomics level,is helpful to enrich the understanding of skin function and understand the mechanism of animal hunger tolerance.
