Spirulina,which is widely used in the field of medical and health care field,contains algae polysaccharide,phycocyanin,(3-carotene,chlorophyll and lutein and other bioactive substances.The products of Spirulina in domestic are mainly spirulina powder,and a small amount of phycocyanin and algae polysaccharide crude extract.However,not only the preparation process is traditional and the raw materials are not fully utilized,but also the product purity is lower.In this study,HPLC-DAD fingerprints was used to analyze the photosynthetic pigments of Spirulina platensis.Four kinds of spirulina photosynthetic pigments were obtained by stepwise extraction of different polar solvents.The main photosynthetic pigments of Spirulina platensis were isolated and purified,Which can lay the foundation for the development and utilization of photosynthetic components and photosynthesis of spirulina.Meanwhile,it can provide guidance for the comprehensive development and utilization of spirulina bioactive components,extend the industrial chain of spirulina processing,and fully improve the utilization rate and economic value of raw materials.The main results of this work are as follows:1 HPLC-DAD fingerprinting method was established to analyze the composition of liposoluble photo synthetic pigments in Spirulina platensis.Spirulina acetone extract mainly contains 23 lipid-soluble photosynthetic pigment components,accounting for 97.52%of the total peak area.After spectral and mass spectrometry analysis,17 kinds of photosynthetic pigments were identified.Among them,there are four kinds of chlorophyll species,and the two kinds of chlorophyll are Hydroxychorophyll a and Chorophyll a.It Contains 8 kinds of chlorophyll derivatives,and the typical structure is Pheophytin a.Besides,there are also five kinds of carotenoids,which are Myxoxanthophyll,Zeaxanthin,Lutein,Canthaxanthin and,?-Carotene.2 The process line of spirulina photosynthetic pigment comprehensive extraction has been established.According to the difference of the polarities of different photosynthetic pigments(?-carotene,lutein,chlorophyll and phycobiliprotein)contained in Spirulina,four crude extracts of photosynthetic pigments were obtained by different polar solvents.After HPLC-DAD detection,the main components were?-carotene,lutein,chlorophyll and phycocyanin.The extraction process of different photo synthetic pigments was respectively optimized by single factor method and response surface method.3 Four high purity photo synthetic pigment monomer were isolated and purified.The separation and purification of P-carotene,lutein and chlorophyll crude were carried out by silica gel column chromatography,alumina column chromatography and recrystallization,and the structure of photosynthetic pigments was identifided by MS,IR and NMR.The results showed that the four high purity pigment monomers were:?-carotene,Lutein,Myxoxanthophyll and Chorophylla.
