Mulberry leaves were the leaf of Morus alba L which were widly cultivated throughout China in most provinces.1-Deoxynojirimycin could combine with such as alpha glycosidase,sucrase and lactase so that disaccharide could not be decomposed to be absorbed by monosaccharose.thus by remarkably restraining the acute increase of bloodafter it,it the breakout and of diabetes. Resveratrol has used in health care products and cosmetics field but on the mulberry leaves in the resveratrol study also rare reports.Based on it,this paper just focused reasearch of column chromat ography to purify DNJ and Resveratro and DNJ was purified by recrystal provied the theory basis for the application of Morus alba L.The main research content is as follow:1. The HPLC detects the contents of Resveratro by quantitative analysis. The conditions of determing liquid chromatogram are as follows:chromatogram column Hypersil ODS (150 mm×4.6 mm,5 um),temperature at 30℃,the wavelength of detectorat298nm,samplesize20μL,rateat1mL/min,methanol:water(v/v):0-2.5-6.8-10=3 0:70-64:36-30:70 as mobile phase. In orderto explore scientific technology,we had to choice the best conditions through orthogonal test. This research had some applicable value because it providedscientific foundation for the industrial production of Mulberry leaves.The best extraction technology was investigation. Resveratro was extracted from mulberry leaveas by ethanol heat reflux extraction,through oneway and orthogonal experiment. The optimum parameter of extraction had been determined:the concentration of ethanol is 80%,the multiple of ethanol to material is 1:13, extraction time is 105min, extraction temperation is 60℃.2. To develop and compare two HPLC methods for determination of 1-deoxynojiri mycin (DNJ) in mulberry leaves. The conditions of determing liquid chromatogram are as follows:chromatogram column Hypersil ODS (150 mm×4.6 mm,5μm),temperature at 30℃,the wavelength of detector at 265 nm,sample size 20μL,rate at 0.8 mL/min, methanol:0.1%acetic acid solution(55:45v/v) as mobile phase.3. The best extraction technology was investigation.1-DNJ was extracted from mulberry leaveas by ethanol heat reflux extraction,through oneway and orthogonal experiment. The optimum parameter of extraction had been determined:the concentration of ethanol is 80%,the multiple of ethanol to material is 1:40, extraction time is 120min, extraction temperation is 70℃, by two extractions,the rate of extraction was 79%.4. The purifying technology of DNJ.The study compared the adsorption properties of seven macroporous resin,including D101,D201,HPD400,HPD100,NKA-9,LSI-4,AB-8,and the resinLSI-4 was determined as the best sorbent. The optimum purification condition obtained was that the DNJ crude extract which concentration was 0.48mg/mL and flow rate on desorption was 2 BV/h was successively eluted with 60% ethanol and 60% ethanol. The purity and the yield of DNJ in 60% ethanol eluate was 35.6% and 60% determined by HPLC.5. The high purity DNJ which purity by D370 type resin cation, The optimum purification condition obtained was that the DNJ crude extract which concentration flow rate on desorption was 2 BV/h was successively eluted with 0.5mol/L ammonia water. The purity and the yield of DNJ in 60% ethanol eluate was 80% and 61.3% determined by HPLC.
