A total of 16 Cellulose decomposition bacteria were separated from carboxymethyl cellulose solid medium and Hutchison filter paper liquid medium.Four strains of cellulose decomposing bacteria with diameter greater than 2.5 cm were screened by Congo red staining.Respectively,Helong-F4,Helong-E6,Helong-A9 and Yanda-L and other strains.CMC enzyme activity of the results of Helong-F4 and Yanda-L strains of CMC enzyme activity than the other two strains more than 2 times.The activity of Helong-F4 +Yanda-L mixed strain was the highest in the single strain and the mixed strain,and its activity was 32.57?mol/mL.According to the 16srDNA gene sequence identification and phylogenetic analysis results,combined with the morphological characteristics of the strain,physiological and biochemical tests,The cellulolytic strain Yanda-L was initially identified as Bacillus subtlis,and the cellulolytic strain Helong-F4 was Btreptomyces badius.The results of the fermentation of the Helong-F4 strain,Yanda-L strain and Helong-F4 + Yanda-L strains were optimized:250mL conical bottled liquid volume of 100mL,the best inoculation amount of 5%-7%by volume inoculation,the most appropriate shaking the number of revolutions 180r/min-200r/min,the optimal incubation time of 48h-72h,the best Culture temperature is30?-35 ?,the optimum initial pH = 7;The optimum carbon source for Helong-F4 and Helong-F4 + Yanda-L strains was glucose,the most suitable carbon source for the Yanda-L strain was sucrose;The optimum nitrogen source for Helong-F4 strain was peptone,and the optimum nitrogen source for Yanda-L strain and Helong-F4 +Yanda-L was beef extract.The degradation rate of corn stalk treated by cellulose decomposing bacteria,the degradation rate of hemicellulose,cellulose and lignin was significantly higher than that of control,ang the degradation rate of corn stalk powder changed with time.In the single strain,the weight loss rate,hemicellulose degradation rate and lignin degradation rate of corn straw powder treated with Yanda-L strain were higher than that of Helong-F4 strain 4%,4.24%and 2.79%respectively,and the degradation rate of cellulose was less than 2.53%of Helong-F4 strain.The degradation rate,cellulose degradation rate and lignin degradation rate of corn stalk powder were higher than that of single strain.Cellulite decomposing bacteria did not significantly inhibit or promote the seed germination of Suzi and Rape seeds.Sterile soil and no sterile soil after adding corn stalk powder,the activity of SOD,POD,CAT and chlorophyll in the leaves of Suzi and Rape treated with cellulose decomposing bacteria were significantly higher than those in control,and Suzi and rape growth index,yield indicators were significantly higher than the control.In different treatments,the activity of SOD,POD,CAT and chlorophyll and the growth index of Suzi and Rape were significantly higher than those of Helong-F4 strain or Yanda-L strain treated with Helong-F4 +Yanda-L strains.
