As a typical symbiotic in degrading cellulose in nature,the symbiotic relationship between termites and microorganisms are fascinating topics for researchers in the world in recent years.To understand the structure of the microbial community,the functions,and the collaboration mechanism of the microbial community,and to figure out the new genes related to degradation of lignocellulose,as well as the goal of establishment of the microbial bionic system,all these can do positive meaning for development and utilization of biomass energy which is vast reserves in the world.In this paper,three groups of Tsaitermes ampliceps which is a forest injurious insect and unique in henan province,were feed on three kinds of food(wood,cornstalk and filter paper)with different content of cellulose.Compare the changes of dominant bacterial community after two weeks,to speculate the possible functions of different bacteria in the termite gut.In addition,A BAC library was constructed as a powerful platform for further study on termite gut microbial.1.There are 4 kinds of Bacteroides,6 kinds of Proteobacteria,3 kinds of Spirochetes,2 Firmicutes and 1 Deferribacleres discovered from Tsaitermes ampliceps by DGGE.The Proteobacteria is highest both because of the number and the abundance.2.The number of advantage bacterium group increased by increasing of the content of cellulose.The group feed with filter paper get the highest number with the largest Shannon index.It indicate that cellulose enriched some microbes in Tsaitermes ampliceps.3.It is found that the different constituents of the diet materials influenced the diversity of gut microbes and their relative abundances in T.ampliceps.The changes in microbiota induced by a 2-week diet shift suggest that Bacteroideres and Spirochaetes9 rather than Firmicutes,digest celluloses,whereas Proteobacteria constantly provide energy,balancing the gut environment.4.It is the first successful genome BAC library construction of termite gut microbes in the world.The library contains 14976 clones,and the average insertion fragment size is 71.7kb with 1075.5Mb storage capacity.The empty loading rate is about 1.04%,and the stability testes results stable.It is a representative library with about all the gene information of enteric microorganism in Tsaitermes ampliceps.
