Termites as a social insect,with strong survival ability,group life long,hidden life and harm a wide range of features,which can cause serious harm to the urban construction,transportation,telecommunications,material storage,fruit trees and other.Termites with wood,weeds or humus for food,which is closely related with the special digestive physiology.Therefore,the digestive system structure,a total of biological flora and in the digestive nutritional of termite has been widely concerned.At present,Study on the digestive physiology of termite mainly concentrated in the digestive tract structure and function,gastrointestinal symbiotic biological in vitro and symbiotic fungi(termites cultivating fungi,fungus garden)and termites digestion,intestinal digestive enzyme separation purification.But termites how to disrupt the wood structure of the lignocellulose,and will be difficult to digest the lignocellulose conversion for its nutritional ingredients.So as to maintain the population growth and breeding,there are still many problems to be further study.This study's research object was Tsaitermes ampliceps in Henan.In this research group,the anatomic structure and the main structures of microbial community were studied,and the two-dimensional electrophoresis method of the contents protein of the termites was constructed.We could find out the differences in protein points through PDQuest software and the chosen points were sequenced by MALDI-TOF/MS,then we could the in-depth analysis the sequencing results through Uni Prot,Ex PASy,NCBI,Gen Bank database.The research could be further studiedon the digestive and metabolic mechanism of termites.The main research contents and conclusions were as follows:1)This experiment had explored the protein extraction and purification methods mainly on the amount of protein,reagents and filtering material of termites gut contents of in Two-dimensional electrophoresis.Finally,we determined 2400 ?g sample quantity,Tris-balanced phenol as the main reagent and the Micro PES resin filter protein.2)We checked the high expression and differences of 39 protein spots by MALDI-TOF / MS sequencing,through the Uni Prot,Ex PASY,NCBI databaseanalysis and prediction the promesenteron and hindgut differences in protein abundance contrast.We found the differential protein spots had 13 in the termite structural protein;they had 9 in the regulating protein;they had 10 in the termite metabolism protein;they had 7 in the gut microbial metabolism protein.We could judge endogenous proteins more than exogenous microbial proteins in the kind and number of proteins.3)We analyze 39 proteins the by the PDQuest software,which had 11 in the promesenteron and hindgut were highly expressed;which had 12 only in the promesenteron expressed,they mainly were the termite metabolism protein and regulatory protein;which had 8 only in the hindgut expressed,they mainly were the microbial metabolism protein;which had 5 in the expression of the promesenteron than hindgut 2 times,they mainly were structural protein;which had 3 in the expression of hindgut greater than promesenteron 2 times.We could speculate the promesenteron of termite mainly proceed digestive metabolism,hindgut mainly proceed microbial metabolic activities.4)In the studies,they had 5 in lignocellulose degrading enzyme involved,the No.3 and No.4 protein which belongs to the endogenous cellulase by the termite itself;The No.5 protein which belongs to the endo-beta 1,4-glucanase by fungi and bacteria;The No.45 protein which belongs to the glycosyl hydrolase 11 th family by the native animals;The No.46 protein which belongs to the Superoxide dismutase by microorganisms.The first three highly had expressed only in the anterior midgut,after two only in the hindgut high expression.We speculated that the degradation of termite of lignocellulose food mainly in the promesenteron,the product further degradation by microbial growth metabolism in the hindgut,and they could provide nutrients Anal feed to other termites.
