Lycium barbarum is a traditional Chinese medicine and health food,and contents L.barbarum polysaccharide(LBP),crude fat and flavonoids and minerals which are necessary for human body as active components.Its functional characteristics are widely accepted by consumers.Lycium barbarum is natural food,more and more people like it.In order to further develop the industry of Lycium barbarum and the quality analysis,the development of quick analysis method is quite important.Fourier transform Infrared(FTIR)spectroscopy is widely used in agricultural products detection with advantages of accuracy,simple operation,and no reagents.The paper used Lycium barbarum which come from different regions in China as raw materials.The regions are classified as Zhongning Lycium barbarum,Non-Zhongning Lycium barbarum and Non Ningxia Lycium barbarum.LBP,crude fat content and total flavonoids were analyzed by chemical methods.FTIR technology combined with chemometrics methods were employed to develop the quantitative calibrations of LBP,crude fat content and total flavonoids.The main research results were as follows:LBP optimal model was developed with pretreatment spectra of multiple scatter correction(MSC),spectral regions 1250-760cm-1.The determination coefficients of calibration(RC2)and validation(RV2)sets were 0.9759 and 09858 respectively when PCs is 5.High linear correlation blind validation predicted and the measured values decision coefficient is 0.9984,the standard deviation is 0.0405.LBP rapid determination method based on FTIR technology with the traditional chemical method were no significant difference.It demonstrates FTIR detection of LBP is workable.Crude fat content PLS calibration was developed with pretreatment spectra of Savitzky-Golay Smothing(S-GS)(2nd,7.3),full spectral regions.The determination coefficients in calibration(RC2)and validation(RV2)sets were 0.9874 and 09894 respectively when PCs is 3.High linear correlation blind validation predicted and the measured values decision coefficient is 0.9890,the standard deviation is0.1061.Total flavonoids content optimal model was developed with pretreatment spectra of MSC with full spectral regions.The determination coefficients in calibration(RC2)and validation(RV2)sets were 0.9950 and 0.9980 respectively when PCs is 7.High linear correlation blind validation predicted and the measured values decision coefficient is 0.9997,the standard deviation is 0.0035.Total flavonoids content determination method based on FTIR technology with the traditional chemical method were no significant difference.It demonstrates the method is workable.
