The purpose of this research is to select perfect variety as research material from special and abundant mangrove resource in Hainan, isolate and identify salt tolerance related protein of mangrove plant quickly. The success of this research would induce more important effect that mangrove resource in Hainan is as germ plasm resource bank and begin a new way for the research and application of salt tolerance of plant.In our subjet, we hope to isolate proteins which expression quantity induce after 500 mmol/L NaCl solution treatment for 12 days, then search homologous protein and obtain it's sequence through MALDI-TOF MS identifying and protein data bank.From 2-D electrophoresis maps, we can find that most of proteins focused between pI5- pI 8 and molecular weight 28KD-66KD. After analysis of 2-D electrophoresis analysissoftware, we find that there are 904 protein spots in the 2-D electrophoresis map whichwas treated with fresh water, 967 protein spots in the 2-D electrophoresis map which wastreated with NaCl solution for 12 days and 790 protein pairs base on the Gel report. At last,fifteen proteins which expression quantity induced after NaCl solution treatment wereselected with histograms on protein groups and method of comparing protein groups fromgel graphs directly, to identify with MALDI-TOF-MS, and then identified results weresearch in protein data bank. It indicates that the Mowse score is 80 between 17# sampleand (CATA3SOYBN) Catalase 3, greater than 66 is significant, molecular weight and pIare adjacent relatively. We can consider 17# sample is (CATA3_SOYBN) Catalase 3; theMowse score is 191 between 29# sample and (ATPB_CUSRE) ATP synthase beta chain,greater than 66 is significant, molecular weight and pI are adjacent relatively. We canconsider 29# sample is (ATPBCUSRE)ATP synthase beta chain; The peptides molecularweight of 53# sample are same with that of 43#, 45#, 52# sample and these samples shouldbe same protein, the Mowse score is 78 between 53# sample and (RCA_ARATH) Spliceisoform Short, greater than 66 is significant, We can consider 53# sample is(RCAARATH) Splice isoform Short. However, (RCAARATH)Splice isoform Shortcan' t effect the salt tolerance ability based on it' s function. MALDI-TOF MS results of49#, 63#, 64#, 66#, 85#, 87#, 88# and 94# sample can' t get reliable information inthe protein data bank.
