The Preliminary Study Of The Protein GCN20 (General Control Non-derepressible-20) In Arabidopsis

Under the condition of nature,different geographical location,changeful climate conditions and human activities,makes the environment complicated and changeable,forming the normal growth and development of plant could bear bad environment,that called adversity or stress.In general,adversity including biotic stress(bacteria,fungi,viruses,pests and weeds,etc.)and abiotic stress(high salt,drought,low temperature,heavy metal pollution,etc.).In eukaryotes,lack of amino acids activated the expression of many genes including downstream of amino acid synthesis and related genes,the control process is mainly contains related proteins GCN1(general control non-derepressible-1),GCN2,GCN20,GCN4.In yeast,the study found that GCN20 is an ABC(ATP binding cassette)family of proteins,its N-terminal combined with GCN1 to form complex,and both have an EF3-like area,to response to the lack of amino acids in organism.With EMS mutagenesis screening drought resistant mutants lew1(leaf wilting mutant 1)in Arabidopsis,we screened the reply mutant sol1-1(Suppressor Of lew1-1).By map-based cloning results,confirmed that At1g64790 gene in sol1-1mutations,according to the bioinformatics analysis,its coding protein SOL1 and protein GCN1 are homologous,and protein GCN1 are involved in the response mechanism of amino acid lacking in yeast cells,mammalian.In plants,the research of proteins related with GCN is relatively small,and no relevant reports if GCN1 /GCN20 complex response amino acid lacking.This topic selected GCN20 as the research object,found a phenotype of gcn20 mutant seedlings that middle of the leaf turn yellow.Through gene cloning methods the full length sequences and promoter of GCN20 were get,respectively linked with 1300-GFP expression vector,1391 carrier,and infected arabidopsis,to study the related function and the response to abiotic stress about protein GCN20.
