Alpine plants are been researched in recent years.All have the unique morphological structure and physiological processes from other general plants because of their special habitats.Chorispora bungeana is highly capable of resisting freezing,drought and ultraviolet radiation,Therefore,it is a ideal species for stress-related genes cloning and researching on the mechanism of stress resistance.CbABF1,a cold-induced gene,is screened out by the method of suppression subtractive hybridization.Our project studied the function of CbABFl from the gene expression patterns,subcellular localization and resistance analysis.We got the following results:1.A whole sequence of CbABFl was isolated from Chorispora bungeana.The full-length sequence of CbABFl is 1482bp,encoding 393 amino acids,the homology of CbABF1 and AtABFl was as high as 71%.CbABFI has 6 bZIP domain and multiple phosphorylation sites.2.Expression pattern of CbABFl is researched By fluorescence quantitative PCR technolo-gy.The results show that CbABF1 gene responses to low temperatures and high salt stresss quickly,and can also be induced by exogenous ABA.So it may be involved in regulating the expression of the downstream gene in ABA signaling pathway.3.From the transcript and protein levels confirmed:CbABF1 distributed among root,stipe and leaf,particularly in the root.Subcellular localization of CbABFl confirmed that the CbABF1 was localized on the cell membrane and nuclear membrane.4.Resistance analysis of transgenic plants displays that CbABF1 gene can significantly improve the frost resistance of tobacco,and reduce the leaf water loss rate.At same time,Overexpression of the CbABF1 in Arabidops is also enhanced the resistance.In summary,CbABF1 gene regulated diverse mechanisms of stress resilience.ABA,low temperature and NaCl induced the expression of CbABFl gene,transgenic plants overexpress the CbABFl gene and will have a higher cold,drought and salinity resistance.This discovery not only enriches the gene pool in Chorispora bungeana,but also contributes to the study of plant stress tolerance mechanisms in the future.
