| Experimental purposes:Explore mi RNA to obesity testicular tissue of rats induced by high-fat diet control and aerobic exercise was revealed and testicular tissue of male high fat diet rats mi RNA interference effect.Methods:The 48 healthy male SD rats were randomly divided into 4 groups:normal diet group(A),(B)A high-fat diet group,the common diet plus exercise group(C),high fat diet plus exercise group(D),each group of 12;Group A and group D,ordinary feed;Group B and group C,high-fat,high-calorie feed.In group C and group D 10 weeks in rats 60 min without weight loading swimming movement,six days a week.Rats after the experiment,measuring weight and fat,testosterone weeks between groups of rats after the experiment quantity each can be divided into 3 groups:group A only 8,11 group B,group C only 10,11 D group,and A total of 12 groups of c DNA library building,use of Solexa high-throughput sequencing technologies combined with biological information analysis appraisal mi RNA and differentially expressed spectrum analysis,further select some differences mi RNA,analyze its biological function.Results:1.Through Solexa sequencing technology,the success rat gene library was constructed,tested for each group of high quality pure sequence(the Clean Data)are more than 20 m,are accounted for over 87%of the original Data,Q30 were above 95%.Pure sequences are classified,in library for containing the mi RNA Unannotated reads.And the small RNA sequences of pure positioning to rat genome,for follow-up study.2.Through biological analysis,464 mi RNAs were identified from group A,group B,group C,and D,and 315 were known mi RNAs and 149 new mi RNAs.According to |log2(FC)|>≥1,the expression of the mi RNA differential expression of the library sequencing is shown in the analysis:the group B has 27 distinct mi RNAs in group A,3 of which are up,and 24 are lower.The C group showed 11 of the mi RNAs in group A,0 of which were raised and 11 were lowered.The D group showed 62 different mi RNAs for group A,37 of which were raised,and 25 were lower.3.According to |log2(FC)|≥1 more points in group A and B mi RNA expression differences,group B to group A differentially expressed mi RNA has 27,three of them,24,predicted the 19612 target genes,of which 24 cut mi RNAs:rno-miR-101a-5p,57.rno-miR-101a-3p,4;rno-mi R-135b-3p,465;Three increases:rno-miR-130b-5p,164;rno-miR-346,2963;Unconservative63922464,3220.4.GO classification shows that the control group and high-fat diet group B group differences of mi RNA genes,and their corresponding target genes in metabolism,reproduction,cell death,the immune system and other aspects have been expression;In KEGG Pathways in the classification,the target genes differentially expressed mi RNA in Pathways in cancer,MAPK signaling pathway and Wnt signaling pathway,Axon guidance,Apoptosis expression.5.By comparison with NR,swiss-prot,and pin-database,the mi RNAs that are associated with sexual reproduction include rno-miR-6328;rno-miR-196b-5p;rno-miR-346;unconservative63922464;rno-miR-503-5p.The mi RNA associated with apoptosis of the cell is rno-miR-6328;mo-miR-3075;rno-miR-346;rno-miR-130b-5p;rno-miR-205;rno-miR-31a-5p;rno-miR-503-5p;unconservative1540112;unconservative63922464.Further,the above mi RNA was apoptosied by Bcl and Bax.Conclusion:1.The success of the experiment through high-throughput sequencing method for common dietary rats,rats a high-fat diet,regular exercise group and high-fat group of testicular mi RNA c DNA library.First has carried on the micro RNA to testicular tissue of rats omics analysis,identified 464 mi RNA,315 of which are known and 149 for the new mi mi RNA RNA.2.More mi RNA in high-fat diet rats testicles cut trend,after aerobic intervention,ordinary group more mi RNA is raised,in short,the difference of mi RNA,all express the amount of change after aerobic exercise,which changes its biological function.3.The mi RNA genes in group A and the high-fat diet group B were associated with sexual reproduction and cell death,according to the classification in the GO and KEGG pathways.4.By comparing with NR,swiss-prot,and pin-database,we believe that the high-fat diet causes the fat rat reproductive cells to be associated with the Bcl,the mi RNA associated with apoptosis of Bax,and rno-miR-6328;rno-miR-3075;rno-miR-130b-5p;rno-miR-346;rno-miR-503-5p;rno-miR-205;rno-miR-31a-5p;unconservative63922464;unconservative1540112;The mi RNAs associated with sexual reproduction are:unconservative63922464;rno-miR-196b-5p;rno-miR-6328;rno-miR-346;rno-miR-503-5p And aerobic exercise intervention can change the expression of these differentially expressed genes,and play an effective improvement of spermatogenic cell death.It is proved that.aerobic exercise is one of the effective methods to improve male infertility. |
