Objective In study of the relationship between different level exercise and the impact on the reproductive function of rats, the mode of aerobic exercise and intensive training was built and the expression of UCP2and the free radical metabolism were observed after swimming with different length of time.Methods24SD male rats were randomly divided into3groups:control group(C),60min swimming group(E6o) and120min swimming group(E12o). The rats in C group was not arranged to swim and that in E6o and E120groups were arranged to swim for10weeks. The level of SOD, MDA and NO in serum was detected, the morphological structure of the testis was observed with hematoxylin-eosin staining, the apoptosis of testicular germ cell was analyzed with TUNEL Method, the apoptosis of sperm was observed with Flow Cytometer and the expression of Cyt C and UCP2was detected after10weeks’ training.Results1. There was no significant difference between the weight of different groups before training and, with training for10weeks, that of E60and E120groups was significant less than that of C group. However only the growth rate of E120group has significant difference with C group.2.The testicular seminiferous tubule of C group was normal, that of E60group had wider interval and mature sperm was visible and that of E120group had damage and the tight junctions had obviously disappeared. The apoptosis sperm of E120group was significantly higher than that of C and E60groups.3.The total sperm number of E120group was significantly less than that of C group and there was no significant difference between E60and C, E120groups. The rate of sperm deformation of E120group was significantly higher than that of C and E60groups, however the sperm activate rate among different groups was no significantly difference.4. The level of GnRH was significantly higher than that of C group and that of E120group was significant lower than that of C group highly significant lower than that of E60group. The concentration of FSH, LH and T in E60group was highly significant different with C group and that of E120group was significant lower than C group, highly significant lower than E60group.5. The concentration of SOD in E60group was significantly higher than that of C group, that of E120group was significant lower than that of C and E60group. The concentration of MDA in group was significantly lower than that of C group, that of E120group was significantly higher than that of C and E60group. The concentration of NO in E60group was significant higher than that of C group and that of E120had significant difference with C and had highly significant higher than that of E60group.6.The expression of Cyt C in E60group was significant lower than that of C group and that of E120was significant higher than that of C and E60group. The expression of UCP2in E120group was highly significant higher than that of C and significant higher than that of E60group.Conclusion1.Intensive training could increase the rate of germ cell apoptosis compared with aerobic exercise for a long time, which hinted that the intensive exercise might be the physiological mechanism that decreased the reproductive function.2. Long-term and appropriate exercise could change the adaptability of antioxidant enzymes which improved the antioxidant capacity of the body.3. UCP2took part in regulating the reactive oxygen and protected body cells from the damage of reactive oxygen which helped to decrease the cell apoptosis.
