Eccentric Exercise On Rat Liver Cell Apoptosis, Regulation Of Gene Bax And Bcl-2 Timing Impact

This study attempts to explore the eccentric exercise on rat liver cell apoptosis and apoptosis-related genes Bax and Bcl-2 the timing of the situation, in order to eccentric training practice in sports science and applications research data.Methods:Healthy male 8-week-old (Sprague-Dawley, SD) rats were 50, were randomly divided into five groups of ten:A quiet control group, exercise group (B1 immediately after exercise group, B2 after exercise 3h groups, B3 and B4 groups 24h after exercise after exercise 48h). The normal control group did not exercise; exercise rats to 16m/min speed, gradient of-16°, continuous training to exhaustion. After the execution of fixed-embedded liver tissue obtained fresh, using TUNEL method in the optical microscope observation of cell morphology and apoptosis in the positioning of the nucleus, quantitative analysis, using immunohistochemical staining and microscopic image analysis system, Bax and Bcl-2 position and positive expression of quantitative analysis.Results:(1) showed that, with the A group compared to the number of eccentric exercise group significantly increased apoptosis, with significant (P<0.01 or P<0.05); the recovery period after exercise at different times, there timing changes, immediately after exercise, when apoptosis of the largest and B1 compared to, B2, B3 and B4 were significantly decreased, with a very significant (P<0.01), but 24h after exercise, the apoptotic index increased after exercise, when the 48h Apoptosis tends to occur near the resting level;(2) The results showed that, compared with A, centrifugal recovery time after exercise in different groups of Bax protein expression increased significantly, with a significant (P<0.05), in which the most obvious expression of B1 group; compared with the B1, With the emergence of post-exercise recovery time is reduced timing, B2, B3 and B4 groups decreased Bax protein expression, with a very significant (P<0.01); B2 and B3 groups compared with little change, there is no statistically significant (P> 0.05); but B4 than B3 significantly decreased (P<0.01), tend to be quiet when the expression level of rat liver cells. (3) The results showed that, compared with A, centrifugal recovery time after exercise in different groups of Bcl-2 protein expression increased significantly, with a significant (P<0.01); with B1 compared, B2 and B3 groups Bcl-2 no significant changes in protein expression, there is no statistically significant (P> 0.05), but the B4 group Bcl-2 protein expression significantly decreased in comparison with the B1 has a very significant (P<0.01). (4) showed that, compared with A, centrifugal exercise group Bax/Bcl-2 ratio was higher than A group, including B1, B2 and B3 increased significantly, with a significant (P<0.05); and B1 compared, B2, B3 and B4 groups Bax/Bcl-2 ratio was significantly decreased, with significant (P<0.05), in which B2 Group B3 group than there is the increased Bax/Bcl-2 ratio; B4 group than any other eccentric exercise groups significantly decreased (P<0.05), tend to be close to A group of Bax/Bcl-2 ratio.Conclusion:The exhaustive eccentric exercise-induced hepatocyte apoptosis, apoptosis is most evident immediately after exercise, with the extension of recovery time, there timing changes; centrifugal 24h after exercise, when this time with the centrifugal delayed soreness after exercise, by the performance of "secondary damage" consistent with the time, the occurrence of apoptosis has been recovered; centrifugal 48h post-exercise body to resume full, the occurrence of apoptosis tends to quiet normal levels.
