Regeneration Across Cold Preserved Peripheral Nerve Allografts Stored In The University Of Wisconsin Cold Storage

Objictive : To investigate the regeneration of peripheral nerve allografts stored in the university of Wisconsin cold storage with different dencity of glutathione.Methods: 80 male SD rats in this experiment were carried out randomly in 8 groups.In group A, the grafts were fresh allografts.In group B,the grafts were fresh autogrsfts.In group C, the allografts were stored in UW solution for 2 weeks.In group D,the allografts were stored in UW solution with 3mol/L glutathione for 2 week.In group E,the allogrsfts were stored in UW solution with 6mol/L glutathione for 2 weeks. In group F, the allografts were stored in UW solution for 4 weeks.In group G, the allografts were stored in UW solution with 3mol/L glutathione for 4 weeks. In group H, the allografts were stored in UW solution with 6mol/L glutathuione for 4 weeks.At 2 week post-engraftment, two grsfts of transplanted nerves were harvested randomly and examined immunohistochemistry. 12 weeks later, the regeneration of sciatic nerve was examined with histology, electro-physiology and auxiliary fibers counted. Result: In the grsfts of nerves harvested at 2 week post-engraftment, the infiltration of lymphocytes in group A was the most evident. group C was inferior, the rest won,t evident. There were a lot of collagenoblast in group A and C, the rest were a lot of Schwann cells. At 12 week post-engraftment, from the histology, electro-physiology and auxiliary fibers counted showed that the nerve regeneration of group B was much better than group D,E,F,G and H, group C was more inferior ,while group A was the worst.Conclusion: 1. The peripheral nerve allografts stored in the University of Wisconsin cold storage were deacresed immunogenicity.2. Increasing the dincity of glutathione in UW solution can prolong the allofrafts.
