ObjectiveBased on the theory of traditional chinese medicine and the theory of active com ponents alignment,the research studies the purgative components of Xiaochengqi decoc tion(Treatise on Febrile Diseases),and analyzes the variable pattern of active compon ents alignment from the compatibility pieces and compatibility doses.The law of chan ge is expected to provide a basis for clinical rational drug use,and to provide referen ce for establishing a quality assessment standard for Chinese medicine corresponding t o clinical efficacy.Methods and contents(1)MethodsTaking the Xiaochengqi decoction(Treatise on Febrile Diseases)as the research o bject and consulting the literature,we sort out the chemical constituents and pharmaco logical activities of XCQD and its compatibility pieces corresponding to the function and indication of XCQD.According to the principles of classical prescriptions,dosage form,solubility and clinical efficacy correspondence,we select purgative ACA of XC QD and its compatibility pieces.Free anthraquinones(aloeemodin-rhein-emodin-chrysoph anol-physcion),conjugated anthraquinones(sennosideA-sennosideB),flavonoid glycosides(na rirutin-narigin-hesperidin-neohesperidin),lignans(honokiol-magnolo).HPLC-DAD four wave lengths were used to determine contents of ACA.?free anthraquinones:chromatographic column for Syncronis C18(250 mm×4.6 m m,5 ?m),mobile phase:methanol-0.1%phosphoric acid,gradient elution,volume flow r ate:0.8ml/min,column temperature:30? sample quantity:10?L,detection wavelengths:254nm.?conjugated anthraquinones:chromatographic column for Syncronis C18(250 mm×4.6 mm,5 ?m),mobile phase:acetonitrile-0.05%phosphoric acid,gradient elution,colu mn temperature:30? sample quantity:10?L,detection wavelengths:340nm.?flavonoid glycosides:chromatographic column for Syncronis C18(250 mm×4.6 mm,5 ?m),mobile phase:acetonitrile-water,gradient elution,volume flow rate:0.7ml/mi n,column temperature:40? sample quantity:10?L,detection wavelengths:283nm.?lignans:chromatographic column for Syncronis C18(250 mm×4.6 mm,5 ?m),m obile phase:acetonitrile-0.05%phosphoric acid;gradient elution,volume flow rate:0.7ml/min,column temperature:30?,sample quantity:10?L,detection wavelengths:294nm.(2)The research contents1)Effects of compatible pieces on ACA of Xiaochengqi dection:?Analysis of Xi aochengqi decoction and its compatibility pieces;?Effect of Rhubarb,Fructus Aurantii Immaturus and Magnoliae Cortex on the ACA of Xiaochengqi Decoction;2)Influence of compatibility dosage change on the ACA of Xiaochengqi decoction:?Effect of si ngle-factor Rhubarb dosage change on the ACA of Xiaochengqi decoction.?Effect of single-factor Fructus Aurantii Immaturus(fried with wheat bran)dosage change on the ACA of Xiaochengqi decoction.?Effect of single-factor Magnoliae Cortex(stir-heated w ith ginger juice)dosage change on the ACA of Xiaochengqi decoction.Results(1)Effect of compatible pieces on ACA of Xiaochengqi dection?Analysis of Xiaochengqi decoction and its compatibility piecesThe purgative components of Xiaochengqi decoction:sennoside B(0.818±0.0079)-se nnosideA(0.504±0.013)-aloeemodin(0.357±0.009)-rhein(0.425±0.010)-emodin(0.094±0.012)-chrysophanol(0.623±0.008)-physcion(0.161±0.006)-narirutin(4.003±0.013)-naringin(58.401±0.092)-hesperidin(3.034±0.012)-neohesperidin(59.056±0.012)-honokiol(0.665±0.009)-magno lo(0.741±0.008).The sum of seven active components in Rhubarb:Rhubarb-Fructus Aura ntii Immaturus(fried with wheat bran)>Rhubarb-Magnoliae Cortex(stir-heated with ginge r juice)>Xiaochengqi Decoction>Rhubarb.Except for Xiaochengqi decoction group,ther e is a significant difference between the other groups and the Dahuang group(P<0.05).The variational trend of the seven active components in each group is similar,but t he cotents and proportions are different.The sum of four active components in Fructu s Aurantii Immaturus(fried with wheat bran):Magnoliae Cortex(stir-heated with ginger j uice)-Fructus Aurantii Immaturus(fried with wheat bran)>Rhubarb-Fructus Aurantii Imm aturus(fried with wheat bran)>Fructus Aurantii Immaturus(fried with wheat bran)>Xiaoc hengqi Decoction.There is a significant difference between the other groups and the f ried Fructus Aurantii Immaturus(P<0.05),and the variational tendency of the four acti ve components in each group is similar,but the cotents and proportions are different.The sum of two active components in Magnoliae Cortex(stir-heated with ginger juice):Magnoliae Cortex(stir-heated with ginger juice)>Magnoliae Cortex(stir-heated with ginge r juice)-Fructus Aurantii Immaturus(fried with wheat bran)>Rhubarb-Magnoliae Cortex(s tir-heated with ginger juice)>Xiaochengqi decoction.There is a significant difference bet ween the other groups and the Magnoliae Cortex(stir-heated with ginger juice)(P<0.05),and the changed trend of the two active components in each group is similar,but the cotents and proportions are different.? Effect of Rhubarb,Fructus Aurantii Immaturus and Magnoliae Cortex on the A CA of Xiaochengqi DecoctionCompared with Xiaochengqi decoction(Dahuangl2g-Chaozhishi9g-Jianghoupu6g),w hen Dahuang,Zhishi and Houpu pieces change,the total amount of effictive compone nts(mg/g)changes as follows:Dahuang-Zhishi-Jianghoupu>Jiudahuang-Chaozhishi-Jiangh oupu>Shoudahuang-Chaozhishi-Jianghoupu>Dahuangtan-Chaozhishi-Jianghoupu?Xiaochen gqi decoction>Dahuang-Chaozhishi-Houpu.The rates of change are Jiudahuang group(6.561%)?Shoudahuang group(4.222%)?Dahuangtan group(0.118%)?Zhishi group(30.186%)?Houpu group(11.218%).In addition to Dahuangtan group,the total effective co mponents of others groups are significantly changed,the most obvious variation is Zhi shi group.(2)Effect of compatible doses on ACA of Xiaochengqi dection?Effect of single-factor Rhubarb dosage change on the ACA of Xiaochengqi dec octionThe doses of fried Fructus Aurantii Immaturus and Magnoliae Cortex with ginger juice remains unchanged.The doses of Rhubarb were 3g,6g,9g,12g,15g,18g and 21g.Compared with the Rhubarb 12g group,the total amount of ACA of Rhubarb,f ried Fructus Aurantii Immaturus,Magnoliae Cortex with ginger juice and Xiaochengqi decoction in other groups have a significant change except for the total amount of AC A of Rhubarb 15g group-fried Fructus Aurantii Immaturus(P<0.05).The change rates o f ACA of Rhubarb in each group are 62.710%(3g),49.463%(6g),18.645%(9g),59.926%(15g),39.772%(18g),44.869%(21g);The change rates of ACA of fried Fructus Aur antii Immaturus in each group are 1.844%(3g),-4.003%(6g),-9.695%(9g),-0.498%(15 g),-6.931%(18g),5.996%(21g);The change rates of ACA of Magnoliae Cortex with g inger juice in each group are-35.348%(3g),43.457%(6g),-5.690%(9g),53.272%(15g),68.777%(18g),19.274%(21g);The change rates of ACA of Xiaochengqi decoction in each group are 49.543%(3g),23.027%(6g),1.579%(9g),-7.323%(15g),-20.472%(18g),-17.032%(21g).Although there is no significant regulation in the total amount of AC A of Rhubarb,fried Fructus Aurantii Immaturus and Magnoliae Cortex with ginger jui ce,the total amount of Xiaochengqi decoction shows a decreasing trend.?Effect of single-factor fried Fructus Aurantii Immaturus dosage change on the A CA of Xiaochengqi decoctionThe doses of Rhubarb and Magnoliae Cortex with ginger juice remains unchanged.The doses of fried Fructus Aurantii Immaturus were 1g,3g,6g,9g,12g,15g and 18 g.Compared with the fried Fructus Aurantii Immaturus 9g group,the total amount of ACA of Rhubarb,fried Fructus Aurantii Immaturus,Magnoliae Cortex with ginger juic e and Xiaochengqi decoction in other groups have a significant difference except for t he total amount of ACA of fried Fructus Aurantii Immaturus 12g group-Magnoliae Co rtex with ginger juice(P<0.05).The change rates of ACA of Rhubarb in each group are 5.366%(lg),11.033%(3g),41.885%(6g),35.446%(12g),38.967%(15g),73.206%(18 g);The change rates of ACA of fried Fructus Aurantii Immaturus in each group are 38.232%(lg),-22.956%(3g),-0.635%(6g),3.652%(12g),1.754%(15g),2.658%(18g);The change rates of ACA of Magnoliae Cortex with ginger juice in each group are 19.061%(1g),32.077%(3g),35.633%(6g),-0.284%(12g),-7.610%(15g),43.385%(18g);The c hange rates of ACA of Xiaochengqi decoction in each group are-73.178%(lg),-62.620%(3g),-22.297%(6g),24.054%(12g),37.528%(15g),52.909%(18g).Although there is no significant regulation in the total amount of ACA of Rhubarb,fried Fructus Aurant ii Immaturus and Magnoliae Cortex with ginger juice,the total amount of Xiaochengq i decoction shows rising trend.?Effect of single-factor Magnoliae Cortex with ginger juice dosage change on th e ACA of Xiaochengqi decoctionThe doses of Rhubarb and fried Fructus Aurantii Immaturus remains unchanged.The doses of Magnoliae Cortex with ginger juice were 0.3 g,1g,3g,6g,9g,12g,and 15g.Compared with Magnoliae Cortex with ginger juice 6g group,the total amount of ACA of Rhubarb,fried Fructus Aurantii Immaturus,Magnoliae Cortex with ginger j uice and Xiaochengqi decoction in other groups have a significant difference except fo r the total amount of ACA of Jianghoupu 1g,3g,9g group-Chaozhishi and Jianghoup u 9g group-Jianghoupu(P<0.05).The change rates of ACA of Rhubarb in each group are 32.763%(0.3g),22.669%(1g),59.256%(3g),28.504%(9g),-7.129%(12g),-12.810%(15g);The change rates of ACA of fried Fructus Aurantii Immaturus in each group ar e-11.831(0.3g),-2.834%(1g),1.088%(3g),-1.920%(9g),-11.144%(12g),-11.306%(15g);The change rates of ACA of Magnoliae Cortex with ginger juice in each group are 65.220%(0.3g),170.555%(lg),-31.294%(3g),-3.698%(9g),15.932%(12g),-17.070%(15 g);The change rates of ACA of Xiaochengqi decoction in each group are 12.767%(0.3g),19.747%(lg),15.191%(3g),-10.585%(9g),-26.855%(12g),-32.817%(15g).Althou gh there is no significant regulation in the total amount of ACA of Rhubarb,fried Fr uctus Aurantii Immaturus and Magnoliae Cortex with ginger juice,the total amount of Xiaochengqi decoction shows a decreasing trend.Conclusion(1)The establishment of ACA of Xiaochengqitang decoction and analysis methodsThe thirteen active components of Xiaochengqi decoction:aloeemodin,rhein,emo din,chrysophanol,physcion,sennoside B,sennoside A,narirutin,naringin,hesperidin,neohesperidin,honokio,magnolol.The 13 medicinal components are the actual substan ces of ACA based on compatible pieces,dosages and dosage forms of classical prescri ptions,and have specifical and effective characteristics.The analysis methods of thirteen active components of Xiaochengqi decoction:HP LC-DAD four-wavelength assay,The methods have good reproducibility,simple operati on,and strong specificity.(2)Effect of compatible pieces on ACA of Xiaochengqi dection?Analysis of Xiaochengqi decoction and its compatibility piecesWhen pieces,dosages and dosage forms are identical,different combinations of T CM can make ACA change.Therefore we can not be free to break the compatible co mbination in clinical use.The quality of Chinese medicine should be maked with clini cal indicators or active components that can represent its efficacy.At present,the cont ent of one or more chemical composition to evaluate the quality of TCM disengaged from clinical,and it is not scientific.? Effect of Rhubarb,Fructus Aurantii Immaturus and Magnoliae Cortex on the A CA of Xiaochengqi DecoctionWhen the Xiaochengqi decoction compatibility dose fixes,the change of compati ble piece can make ACA vary.The active components of different pieces of a chinese medicine have a difference,and the impact on other pieces in prescription is also dif ferent.They belong to different medicines,and can not be freely replaced in prescripti on.different pieces of a chinese medicine are different drugs,and can not be randoml y used.The quality standards of different pieces should also be maked on the clinical efficacy and active components.(3)Effect of compatible doses on ACA of Xiaochengqi dectionWhen the pieces of XCQD are changeless,the variable doses of Rhubarb,fried F ructus Aurantii Immaturus and Magnoliae Cortex with ginger juice cause changes of A CA of Xiaochengqi decoction.The variable doses make different drugs,and we can n ot change dosage in clinical use.Dose is one of the key factors that affect the ACA of Chinese medicine.In summary,the factors influencing the the purgative components of Xiaochengqi decoction are:pieces,compatibility,dosage.One of these factors changes and the effe ctive component changes.The active components have the basic characteristics of spec ificity and effectivenes.
