Purpose: Osteoarthritis(Osteoarthritis,OA)is one of common clinical orthopedic disease.In articular cartilage degeneration and destruction,the subchondral bone,cystic change,as well as the joint edge and the subchondral bone with osteophyte formation of a nonspecific inflammation.The main clinical manifestations of joint pain,stiffness,dysfunction,is one of the common diseases orthopedics.Osteoarthritis patients also increases in the elderly with population aging.By flavored antlers bolus of papain and cysteine to osteoarthritis in rats of experimental research,observational studies Jiawei Lujiao Mixture(JLM)to treat osteoarthritis and mechanism of action,provide pharmacological basis for clinical application.Material and method:56 male and 56 female Wistar rats were raised in Liaoning University of Traditional Chinese Medicine experiment center offers.Using papain cysteine cause formation of osteoarthritis in rats model,randomly divided into 7 groups,model group,JLM low,medium and high dose group,double vinegar for capsule group,the strong bone joints pill group.Papain 4% and 0.3 M cysteine were used for rat knee articular cavity injection,and the normal group was given the same volume of saline and gavage for 6 weeks.JLM low,medium and high dose group,respectively 5 g/kg/d,15 g/kg/d,50 g/kg/d,and western medicine group was given double vinegar for 0.01 g/kg/d,Chinese traditional medicine control group was given strong bone joints,1.2 g/kg/d,Take out rat knee joint after the test,and gross and imaging observation,histological examination were performed to evaluate the protect effects of JLM.Collecting the supernatant of femoral artery blood,3000 /min,10 min,and detecting of IL-1 beta,TNF-alpha,PGE2.Results:1.The gross observation of the knee joint surface damage degree scoremodel group was higher than normal group,P < 0.01.JLM low,medium and high dose group compared with model group,all groups had statistical significance,P < 0.01.Strong bone joints pill group and double vinegar group were statistically significant,P < 0.05.2.HE staining scores of the model group was higher than that of normal group,P < 0.01.Compared with model group,JLM high dose group was decreased,P < 0.01 and strong bone joints pill and double vinegar were decreased,P < 0.05,P < 0.01.3.IL-1 ?,TNF-? content of the model group were higher than the normal group which suggested that the experimental model group was success.Compared with model group,flavored antlers medium dose group decreased the content of serum IL-1 ?,P < 0.05.JLM high dose group decreased serum TNF-? content,P < 0.01,compared with model group,and compared with strong bone joints pill group,serum TNF-? content of JLM high dose group decreased,P < 0.05.Conclusion:Animal model was established successfully,light microscopy score showed: JLM showed a protective effect in knee arthritis rats caused by papain and cysteine.The HE score showed,the high dose group and model dose group of modified antler pill have protective effect on rats with knee osteoarthritis induced by papain and cysteine.In addition,the middle dose group of JLM could reduce the expression of IL-1 beta,and reduce the inflammatory reaction of articular cartilage,and protect the normal physiological function of articular cartilage.4.The high dose group of JLM could reduce the expression of TNF-?,and reduce the inflammatory reaction of articular cartilage,and protect the normal physiological function of articular cartilage.The effect is better than Strong bone joints pill group.5.No significant effect on the change of serum PGE2 level in JLM group compared with model group.
