To explore the mechanism of Modified Duhuo Jisheng Mixture on knee osteoarthritis.Methods:1?Analyzing the difference between plasma samples of healthy people and KOA patients by gas chromatography tandem mass spectrometry(GC-MS),and screen for the specific metabolic markers;2 ? KOA patients were intervened with Modified Duhuo Jisheng Mixture,analyzing the changes of biomarkers and the results of the construction of metabolic pathways to explore the related pathways of Modified Duhuo Jisheng Mixture for the treatment of knee osteoarthritis.Results:1?Screening results of specific markers: Compared with the healthy people,the contents of lactic acid,acetic acid,alanine,glycerin,succinic acid,2-keto-isopropylic acid,glucose,ribose acid and galactose in the serum of KOA patients were significantly increased(p < 0.05),and the contents of aminomalonic acid,threonine,proline and inositol were significantly decreased(p < 0.05);2?The effect of Modified Duhuo Jisheng Mixture on the specific markers of knee osteoarthritis: the contents of lactic acid,alanine,glycerin,succinic acid,glucose and arachidonic acid in the serum of KOA patients were significantly increased(p <0.05),and the contents of aminomalonic acid,threonine,proline,and inositol were significantly increased(p < 0.05);3 ? The main metabolic pathways such as citrate cycle,glycerol metabolism,arachidonic acid metabolism,starch and sucrose metabolism,inositol phosphate metabolism were found to play a crucial role in the callback of metabolites.Conclusion:1?Specific markers related to knee osteoarthritis may include: lactic acid,acetic acid,alanine,glycerol,succinic acid,2-ketoisopropionic acid,glucose,ribonic acid,galactose,aminomalonic acid,threonine 14 different markers of acid,proline,inositol content,and arachidonic acid;2 ?
