Purpose: In this experiment, we used D-galactose (D-galactose) to induce senile rats as the research object. The therapy of Spleen-Supplementing Qi-Medicated Medicinal Meals and Acupoint Application on aging model, to observe the changes of skeletal muscle morphology,antioxidant capacity and expression of caspase-3 and caspase-8 in aging rats, understand the intrinsic relationship between the progressive decline of skeletal muscle mass and the ability of apoptosis and anti-oxidation, and the mechanism of action. To provide theoretical basis for the clinical intervention Sarcopenia effective nursing measures to improve the elderly daily activities, self-care ability, reduce the risk of falls and bed, improve the quality of life of the elderly , Reducing health care costs and the economic burden of families and society.Material and method: Thirty-six healthy male Sprague-Dawley rats (SPF) aged 3 months were randomly divided into blank group, model blank group, and diet and acupoint application group, with 12 rats in each group. 7.5% D-gal 125mg / kg was injected subcutaneously into the abdomen subcutaneously in the model blank group and the diet and acupoint application group. The rats in the blank group were injected abdomen subcutaneously with 0.9% normal saline 125mg / kg every day for 6 weeks. After 6 weeks, the diet and acupoint application group was treated with the combination of traditional Chinese medicine (10ml · kg-1 · d-1) and acupoint application, and the blank group and the model blank group were given equal volume of water and vaseline for 4 weeks. 24 hours after the last gavage, abdominal anesthesia, abdominal blood through the abdominal aorta, rapid stripping of the rectus femoris.The morphological changes of rat rectus muscle cells were observed by HE staining. Real-time quantitative polymerase chain reaction (PCR) was used to detect the expression of Cysteinyl aspartate specific proteinase-3 (Caspase-3) and Caspase-8 mRNA in the rectus femoris muscle. The activities of glutathione peroxidase (GSH-Px),superoxide dismutase (SOD) and the content of malondialdehyde (MDA) in the serum and GSH- Px, SOD, Caspase-3 ,Caspase-8 and the content of MDA in the rectus femoris muscle.Statistical analysis software SPSS17.0 and Microsoft Office Excel 2007 were used for data processing analysis. Each group of data was expressed as Mean ± S. The differences between the groups were analyzed by independent samples T test, and the significant difference was defined as P <0.05Results:1. The rats in the model blank group and the group of diet and acupoint application had the characteristics of aging, However, the aging characteristics of rats in the group treated with diet and acupoint application were less than those in the control group. The weight of rats in three groups were increased, rats in the model blank group were slightly lower than those in the blank control group,but the difference was not statistically significant (P> 0.05).2. Under light microscope: the rats in the blank group had normal skeletal muscle histology,and the rats in the model blank group and the group of diet and acupoint application had different degrees of muscle cell atrophy or degeneration.3. The serum GSH-Px and SOD activities of the rats in the model blank group were significantly lower than those in the blank group (P <0.05), and the content of MDA was significantly higher than that in the blank group (P <0.05) ;The activity of GSH-Px and SOD in the serum of the rats in the diet and acupoint application was significantly higher than that in the model blank group (P <0.05) ,and the content of MDA was significantly lower than that in the model blank group (P <0.05).4. Skeletal muscle biochemical indicators: the activities of GSH-Px and SOD in the skeletal muscle of the blank group with model blank group were significantly decreased (P <0.05), the content of MDA and the activities of Caspase-3 and Caspase-8 were significantly increased (P<0.05); Compared with the model blank group, GSH-Px and SOD in skeletal muscle increased significantly (P <0.05), the content of MDA and activity of Caspase-3 and Caspase-8 in the skeletal muscle of rats in the diet and acupoint application group were decreased significantly (P <0.05).5. Skeletal muscle tissue molecular biological indicators: Compared with the blank group, the expression of Caspase-3 and Caspase-8 mRNA in the skeletal muscle of the model blank group increased significantly (P <0.05); Compared with the model blank group, the expressions of Caspase-3 and Caspase-8 mRNA in the skeletal muscle of rats in the diet and acupoint application group were significantly decreased (P <0.05).Conclusion:1. Diet and acupoint application can improve the appearance, morphology and skeletal muscle histology of aged rats, and can reduce the occurrence and development of Sarcopenia.2. Diet and acupoint application can improve the activities of GSH-Px and SOD in the serum and skeletal muscle of ageing rats, decrease the content of MDA, increase the antioxidant capacity of skeletal muscle, reduce the oxidative damage of skeletal muscle and delay the aging of skeletal muscle?3. Diet and acupoint application can decrease the activities of Caspase-3 and Caspase-8 in skeletal muscle cells of aging rats, decrease the expression of Caspase-3 mRNA and Caspase-8 mRNA, and decrease the apoptosis and delay the senescence of skeletal muscle.
