Purpose:To observe the effect of electroacupuncture on TGF-?1/ Smads signaling pathway in rats with prehypertension induced by salt-sensitive prehypertensive rats "Quchi and Zusanli",and to detect the expression of TGF-?1 Smad2 Smad3Smad7?-SMA,and to elucidate the inhibitory effect of acupuncture on the abnormal transformation of renal epithelial cells to myofibroblasts in prehypertensive rats.Material and method: Ten male salt-resistant rats?SS?were set as normal group,and 30 male dahl salt-sensitive rats?DS?were randomized into a model group,Acupuncture group and non-acupoint group?n = 10?.Adaptive feeding for 7 days,no abnormalities after high salt diet?8% Nacl?feeding 12 days.When the blood pressure increased to 120-139/80-89 mm Hg range is the success of modeling,while stopping high-salt feed to feed,to feed the general feed.High salt diet during feeding free water,the daily replacement of sterile litter to ensure a good living environment in rats.Acupuncture group was given electroacupuncture at "Quchi" and "Zusanli" two points?"Zusanli"?located on the lateral side of the knee of the rat,about 5 mm away from the fibular head,"Quchi" in the proximal end of the rat radius?15 mm in bilateral iliac crest,20 mm in posterior?,and a non-acupoint in the non-acupoint group was selected as the control point Point).The acupoints of the two groups of rats were pricked and the needling depth was about 3mm,then the needle was twisted.After about half a minute,the acupuncturing instrument was taken,and the frequency was 2HZ,20 min.Once a day for 6 times a week for a total of 5 weeks,weekly observation record blood pressure in rats 1 times.The m RNA expression of Smad3 Smad7 and ?-SMA in the left kidney of the rats was detected by real-time quantitative PCR?q RT-PCR?and the expression of P-Smad2 was detected by Western Blot Expression level.Results:1.The expression of TGF-?1 Smad2 Smad3 Smad7 and ?-SMA m RNA in the renal tissue of each group:The expression of TGF-?1 in the model group was significantly higher than that in the model group?P <0.05?.The expression of Smad2 gene in the model group was significantly higher than that in the model group?P <0.05?.Smad3 gene expression changes,the expression of each group no significant change.The expression of Smad2 gene in the model group was significantly higher than that in the model group?P<0.05?.The expression of Smad7 gene decreased,compared with the normal group?P<0.05?.Compared with the model group,the expression of Smad7 increased?P<0.05?.The expression of ?-SMA in the model group was significantly higher than that in the model group?P <0.05?.2.The expression of P-Smad2 P-Smad3 protein in the kidney of each group The protein expression of P-Smad2 in the model group was significantly higher than that in the model group?P <0.05?.Conclusion:1.Abnormal activation of TGF-?1 / Smads pathway in renal tissue of salt-sensitive prehypertensive rats.2.Electroacupuncture stimulation "Quchi,Zusanli" two points can reduce the TGF-?1Smad2 and ?-SMA m RNA content,increased m RNA content of Smad7.3.Electroacupuncture stimulation "Quchi,Zusanli" two points can reduce the P-Smad2 protein content.4.The effect of electroacupuncture at "Quchi" and "Zusanli" points on the renal function of rats with salt-sensitive prehypertension may be related to the signal transduction of TGF-?1 / Smads signaling pathway.Expression,inhibition of renal epithelial cells to myofibroblast cells related to play a role in the protection of the kidneys.There was no significant change in the expression of P-Smad3 protein in each group.
