| Purpose:By observing the effect of electro-acupuncture “Zusanli†and “Quchi†points for systolic blood pressure, TNF-a content in the serum and expression of NF-k Bp65,Ik B,Ik K and m RNA in thoracic aorta for prehypertension artery in rat tail, to explore the correlation between Ik K/NF-k B signal pathway and prehypertension,and try to explore the curative effect and action mechanism of acupuncture treatment for prehypertension,to provide scientific experimental data and theoretical basis for prevention and treatment of prehypertension。 Materials and methods:10 salt resistant rats(SS) for 7 week-age as normal group,and 30 Dahl salt sensitive rats for 7 week-age,which were randomly divided into 3 groups,10 rats in each group,namely model group,acupuncture group,non acupoint group。After modeling successfully,each group rats was feed normally,and the normal control group for the salt resistant rats had no treatment; while the model group for salt sensitive rats fixed 20 min daily without acupuncture; acupuncture group selected “Quchi†“Zusanliâ€acupoints in rats according to “experimental acupuncture scienceâ€,and inserting by using filiform needle about 3mm for electro-acupuncture(EA parameters:density wave,frequency 2Hz) intensity was accorded with trembling of needle handle,and stimulationfor 20 min,once a day,6 times per week for treatment,and continuously for 4 weeks; The selection of“Zusanliâ€acupoint in rats for non acupoint group was corresponding points beside the Zusanli1-2mm,corresponding points for“Quchi†was selected in junction of upper arm front deltoid and biceps, and treatment methods and time were same with acupuncture group。Acupuncture treatment ended after 4 weeks,taking out serum, thoracic aorta from rats.Detect the content of TNF-a in rats serum by using ELISA;and NF-k B p65,Ik B,Ik K m RNA expression in thoracic aorta with fluorescence quantitative PCR method。 Results: 1.The effects for rat tail artery systolic pressure with prehypertension by electro-acupuncture at“Zusanliâ€,“Quchi†acupoint。Rat tail artery systolic pressure in the model group was compared with the rats in normal group,which was significantly increased(P<0.01), compared with the model group, rat tail artery systolic pressure in acupuncture group and non acupoint group was significantly decreased(P<0.01), acupuncture group rats were compared with the normal group,which was no statistical significance(P>0.05)。 2.The effects for TNF-a content for prehypertension in rats serum by electroacupuncture at “Zusanliâ€,“Quchi†acupoint。Compared with the rats in model group, contents of TNF-a in rats serum in normal group was significantly reduced(P<0.01), Compared with the rats in model group,the contents of TNF-a in acupuncture group rats was significantly decreased(P<0.01), acupuncture group rats were compared with normal rats,which was no statistical significance(P>0.05),the model group was compared with non acupoint group rats,which was no statistical significance(P>0.05)。 3.The effects for NF-k Bp65 m RNA expression in prehypertension rat thoracic aorta by electro-acupuncture at “Zusanliâ€,“Quchi†acupoint。Compared with the model group, r NF-k Bp65 m RNA expression in prehypertension rat thoracic aorta in normal group were significantly decreased(P<0.01), NF-k Bp65 m RNA expression in rat thoracic aorta in model group was compared with the acupuncture group rats,which was significantly decreased(P<0.01), acupuncture group rats was compared with normal group,which was no statistical significance(P>0.05),rats in the model group was compared with non acupoint group rats,which was no statistical significance(P>0.05)。 4.The effects for Ik Bm RNA expression in prehypertension rat thoracic aorta by electro-acupuncture at “Zusanliâ€,“Quchi†acupoint。Compared with model group,Ik B m RNA expression in rat thoracic aorta in normal group was significantly increased(P<0.01), Ik B m RNA expression in rat thoracic aorta in model group was compared with the acupuncture group rats,which significantly increased(P<0.01),acupuncture group rats was compared with the normal group,which was no statistical significance(P>0.05),the rats in the model group was compared with non acupoint group rats,which was no statistically significant(P>0.05)。 5.The effects for Ik K m RNA expression in prehypertension rat thoracic aorta by electro-acupuncture at “Zusanliâ€,“Quchi†acupoint。Compared with the model group rats, Ik K m RNA expression in rat thoracic aorta in normal group was significantly decreased(P<0.01),and compared with the model group rats,Ik K m RNA expression in rat thoracic aorta in acupuncture group was significantly decreased(P<0.01), acupuncture group rats was compared with normal rats,which was no statistical significance(P>0.05), rats in the model group was compared with non acupoint group rats,which was no statistically significant(P>0.05)。 Conclusion: 1.It has good antihypertensive effect by electro-acupuncture at “Zusanliâ€,“Quchi†acupoint on prehypertension rat。 2. Electro-acupuncture at bilateral “Zusanliâ€,“Quchi†point is possibly regulating the expression of inflammatory factors and inflammatory reaction by inhibiting the activation of the Ik K/NF-k B signaling pathway,thereby down, to further reduce the occurrence and development of prehypertension。... |
