| Background With the improvement of science and technology and the application of radiation in clinical treatment,people are more and more likely to be exposed to rays,which results in the high incidence of testicular spermatogenesis damage or even infertility.The current incidence of male infertility accounts for about 15% to 50% of infertile couples,and the rate is on the rise.And the incidence of reproductive health problems for men with long-term exposure to radiation is higher.Therefore,the impact of radiation on male reproductive system arouse more and more attention.Objective This study aims to explore traditional Chinese medicine(TCM)etiology and pathogenesis of the acute radiation causing testicular injury from the perspective of "renal essence deficiency",to establish the treatment principles of TCM preventing testicular injury caused by acute radiation,to enrich TCM prevention and treatment theory of testicular injury caused by acute radiation,and to establish a safe and effective TCM prescription for preventing testicular radiation injury through experimental study and observation on the prevention and protect effect of TCM prescription on the mice's testicular radiation injury.Method Part one-theoretical study: Search the literature and sort out TCM theories of "kidney essence deficiency" and sum up the clinical manifestations of kidney essence deficiency;discuss the etiology and pathogenesis of testicular injury caused by acute radiation,and improve TCM etiology theory of testicular injury caused by the acute radiation.Part two-experimental study: This study group selected the TCM prescriptions which have certain preventive and protective effect on the whole body radiation injury through previous experiments.This experiment explored the basic preventive and protective effect of TCM prescription on testicular injury caused by acute radiation.In this experiment,male Balb / c mice were used as subjects in the blank group,the model group,the Anduolin group,the high-dose TCM prescription group and the low-dose TCM prescription group.14 days after medication,the mice were treated with 5.5Gy60Co? ray radiation through the whole body,and some were killed and were performed the following experiments 1 day after the radiation,7 days,21 days,and 35 days respectively: Experiment 1: The basic prevention and protection of TCM prescription on mice's testicular injury caused by acute radiation The body weight,testis index,epididymis index and whole blood cells of mice in all groups were measured at four time points.Experiment 2: Study on preventive and protective effect of TCM prescription on mice's sperm injury caused by acute radiation The sperm motility,sperm count,sperm density and sperm velocity of mice in all groups were measured at four time points.Experiment 3: Prevention and protection of TCM prescription on mice's testicular morphological structure caused by acute radiation Prevention and protection of TCM prescription on mice's testicular morphological structure caused by acute radiation was tested through observing the pathological changes of testicular structure of mice in all groups at four time points.Experimental 4: Study on the preventive and protective effect of TCM prescription on mice's androgen level caused by acute radiation The changes of serum sex hormones in the mice of all groups were detected at four time points.Results Part one-theoretical study: In the traditional theory of TCM etiology it hasn't been found that there were records on the testicular injury caused by ionizing radiation,and modern physicians analyzed the etiology and pathogenesis of testicular injury caused by acute radiation according to its clinical manifestation.The discussion of the etiology would provide a certain theoretical basis for TCM prevention of testicular injury caused by acute radiation.Part two-experimental study: 1.The basic prevention and protection of TCM prescription on mice's testicular injury caused by acute radiation Ray was highly lethal.The weight of Balb/c mice decreased rapidly after exposure to 5.5 Gy,and the weight of mice receiving radiation decreased to the lowest point on the 16 th day after irradiation.The testis index and epididymis index were significantly decreased on the 21 st day and 35 th day after radiation.Compared with the model group,the testis index and epididymis index of mice in the high dose group and the low dose group increased on the 21 st day and 35 th day.2.Study on preventive and protective effect of TCM prescription on mice's sperm injury caused by acute radiation The sperm quality of Balb/c mice was not significantly decreased on day 1 and day 7 after radiation with 5.5Gy,and the sperm count and sperm motility were significantly decreased on the 21 st day and 35 th day,so was the sperm velocity.At the later stage of irradiation,the sperm quality of the high dose group and the low dose group increased at different degrees on the 21 st day and 35 th day compared with the model group.3.Prevention and protection of TCM prescription on mice's testicular morphological structure caused by acute radiation Morphological changes of testicular pathological damage after 5.5Gy60Co? ionization radiation continued to aggravate with the passage of time.TCM prescription could effectively protect the pathological morphological changes of testis and epididymis after 21 days and 35 days during the later stage when severe injury occurred.4.Study on the preventive and protective effect of TCM prescription on mice's androgen level caused by acute radiation In the early stage of acute ionizing radiation(1 day and 7 days after radiation),the protective effect of TCM prescription on testosterone and estradiol in mice's serum was not significant;but in the middle and long period after radiation(21 days, 35 days after radiation),the protective effect of TCM prescription on the mice's sperm quality parameters and testosterone and estradiol was more obvious.Conclusion Part one-theoretical study: The occurring of acute radiation-induced testicular injury and its prevention and treatment are closely related with the kidney essence deficiency,so we can develop anti-ionizing radiation TCM prescription to reduce the induced testicular injury from the perspective of ?treatment from the kidney?.And during the medication for prevention,the TCM prescriptions of benefitting the essence,generating the marrow,benefitting qi and nourish yin should be chosen.Part two-experimental study: On the basis of successfully replicating animal model of acute radiation-induced testicular injury,this experiment found that TCM prescription's overall prevention and protection on the mice's testicle was significantly effective 21 days and 35 days after the radiation through the test of mice's testicular base index,sperm quality and function and androgen levels. |
