MicroRNA-192 And -215 Participate In The Study Of Human Gastric Cell Biological Function Through The Target Gene BIVM

MicroRNA(miRNA)is a class of endogenous,non-coding 19-22 nucleotides RNA,discovered in recent years.The“seed region”,which is the core sequensce that enscompasses the first 2-7 nucleotides at the 5'portion of miRNA,is essential for the specific suppression of target mRNA.The first miRNA lin-4 Was discovered in 1 993.Sinsce then,more than 500 miRNAs aberrantly expressed in different types of cancers Was confirmed.Inscreasing numbers of studies showed that miRNAs were involved in the regulation of the proliferation,differentiation and apoptosis.Therefore,miRNAs were deemed to play a crucial role in the initiation and progression of human canscers and act as many onscogenes and turnor suppressor genes.Gastric canscer is one of the most common malignanscies around the world.Altered expressions of miRNAs have been reported in gastric canscel Some of these miRNAsmay play a critical part in carcinogenesis.Now,it has been confirmed that miR-21,miR-191,miR-223,let-7a microRNA,miR-106b-25 subsets related to the occurrensceof gastric canscer,whereas,let-7 microRNA,mi R-155 related to the invasion andmetastasis of gastric canscer.At present,there is no research on the relationship between BIVM(for basic,immunoglobulin-like variable motif-containing)and gastric canscer.We had consulted relevant literature and earlier research on miRNA expressions in gastric canscer,initially,and identified BIVM is the focus of our research.Four commonly databases inscluding microRNA.org,TargetScath Pictar and miRanda miRNAs were used to forecast the miRNAs with the search terms of BIVM.miR-192 and-215 with high predictive values(at least predicted by three databases)were selected as candidate mi Rs;Gene microarrays?real-time RT-PCR or Western blot was used to detect that miR-192 and-215 may be the interacting miRNA of BIVM;Luciferase assays suggested that miR-192 /-215 inhibits activated BIVM expression at the posttranscriptional level,but mi R-192 /-215 mimics play the opposite roles.In gastric cell line HFE145?SGC7901?BGC823 transfecting BIVM-si RNA decreased BIVM protein expression to research the effect of BIVM on cell biology funsction.
