Objective Observe some indexes(body fat Parameters?blood lipid?islet function evaluation and so on)of the peace constitution normal control group?overweight group(non-phlegm-dampness constitution and phlegm-dampness constitution)?phlegm-dampness constitution obesity group,use the study data of proteomics and metabolomic,by Analysising and comparative studying four groups'the characteristic of serum protein group and specific characteristic metabolites,to indicate the patholo-gical basis of the formation of TCM phlegm-dampness constitution,Further clarify TCM phlegm-dampness constitution associated with the pathogennesis of obesity,to provide theoretical basis for TCM phlegm-dampness constitution physical therapy.Methods The study uses high performance liquid chromatography coupled with Q-TOF/MS,and protein fingerprint technology?with serum samples of the peace constitution normal control group?overweight group(non-phlegm-dampness constitution and phlegm-dampness constitution)?phlegm-dampness constitution obesity group,combined with insulin resistance,sugar,lipid metabolism and other indicators,through the comparison and analysis of experiment data,indicate serum metabolic spectrum and differentially expressed protein group.Results the results of analysis and compare the Metabolomics and proteomics mass spectrometry:1.the peace constitution normal control group and non-phlegm-dampness constitution overweight group:(1)Proteomics research:no significant differences in protein;(2)Metabolomics research:Identified five differences serum metabolites:Alpha-N-Phenylacetyl-L-glutamine?PGF2a diethyl amide?3-(Uracil-1-yl)-L-alanine?Propionyl-L-carnitine?and LysoPC(18:3(6Z,9Z,12Z)).2.the peace constitution normal control group and phlegm-dampness constitution overweight group:(1)Proteomics research:no significant differences in protein;(2)Metabolomics research:identified eight differences serum metabolites:acetonide?N-gamma-L-Glutamyl-L-Pheny-lalanine?Decanoyl-L-camitine?L-Arginine?O-octanoyl-R-camitine?N-palmitoyl serine?Phytosphingosine?C17 Sphinganine?3?the peace constitution normal control group and phlegm-dampness constitution obesity group:(1)Proteomics research:identified three kinds of differences in protein:DNA-dependent protein kinase catalytic subunit?FBW1A(F-box/WD repeat-containing protein 1A)?LARGE(Glycosyltrans-ferase-like protein LARGE1);(2)Metabolomics research:identified eight differences serum metabolites:LysoPC(18:3(6Z,9Z,12Z))and LysoPC(18:3(9Z,12Z,15Z))PC(0-18:1(9Z)/0:0)?Propionyl-L-camitine?Alpha-N-Phenylacetyl-L-Glutamine?LysoPE(0:0/22:6(4Z,7Z,10Z,13Z,16Z,19Z))?PE(22:6(4Z,7Z,10Z,13Z,16Z,19Z)/0:0)?PGF2? diethyl amide?4?non-phlegm-dampness constitution overweight group and phlegm-dampness constitution overweight group:(1)Proteomics research:no significant differences in protein;(2)Metabolomics research:identified ten differences serum metabolites:Hydroxyproly1-Tyrosine?3-(Uracil-1-yl)-L-alanine?acetonide?L-Carni-tine?N-Oleoyl-L-Serine?PE(20:4(5Z,8Z,11Z,14Z)/0:0)?N-palmitoyl serine?PS(O-18:0/0:0)?Decanoyl-L-carnitine?LysoPE(20:3(11Z,14Z,17Z)/0:0)?5?phlegm-dampness constitution overweight group and phlegm-dampness constitution obesity group:(1)Proteomics research:no significant differences in protein;(2)Metabolomics research:identified nine differences serum metabolites:LysoPC(18:3(9Z,12Z,15Z))and LysoPC(18:3(6Z,9Z,12Z))?Phytos-Phin-gosine?LysoPE(20:3(11Z,14Z,17Z)/0:0)and LysoPE(0:0/22:6(4Z,7Z,10Z,13Z,16Z,19Z))?acetonide?Alpha-N-Phenylacetyl-L-Glutamine?N-Oleoyl-L-Serine?PC(0-18:1(9Z/0:0))?Conclusion1.In proteomics research,Compared with the normal group,the expression of serum differential protein was found in the phlegm-dampness and non-phlegm-dampness group.Compared with the phlegm-dampness group,the expression of serum differential protein was not found in the non-phlegm-doping group and the phlegm-damp obesity group.Compared with the phlegm-dampness group,the expression of serum differential protein was not found in the non-phlegm-doping group and the phlegm-damp obesity group.2.There were metabolites such as Alpha-N-Phenylacetyl-L-Glutamine and LysoPC(18:3(6Z,9Z,12Z))in the the peace constitution normal control group?phlegm-dampness constitution overweight group?phlegm-dampness constitution obesity group,among them,Phenylacetyl-L-Glutamine in the phlegm-dampness constitution obesity group was higher than that in the peace constitution normal control group and the phlegm-dampness constitution overweight group,LysoPC(18:3(6Z,9Z,12Z))in the phlegm-dampness constitution obesity group was lower than that in the peace constitution normal control group and the phlegm-dampness constitution overweight group.And the concentration of LysoPC(18:3(6Z,9Z,12Z))in the phlegm-damp obesity group was significantly lower than that in the peace constitution normal control group.Suggesting that there is differential expression of serum metabolites in phlegm constitution.3.From the perspective of cure not ill choose metabonomics monitoring is more advantageous to the prevention of obesity.
