Objective: aim to analyze the effects of PRP in combination with TSC on tendinopathy by observing the expression of type I,type ? collagen and MMP-1 in the tendon tissue of rabbit Achilles tendon model,and try to discuss the function mechanism of PRP in combination with TSC treating tendinopathy.At the meantime,we try to seek the new thinking and safe treatment of tendinopathy and try to provide theoretical basis for its clinical application.Methods: Forty New Zealand white rabbits were randomly divided into model group(n=32)and blank control group(n=8).In the model group,the animals were injected with PGE2300 ng /0.2ml,1 time per week,and the animal model of rabbit Achilles tendinopathy was established.The rabbits in blank control group were injected equal amount of medical saline.After 4 weeks,8 rabbits were randomly selected from the model group to compared with the blank control group under musculoskeletal ultrasound to analyze the model effect.Then,model rabbits were randomly divided into 4 groups again: A group(the blending group),B group(TSC group),C group(PRP group)and D model control group,each group of 8 rabbits.When the animal model was stable,the blends(group A)were injected PRP and TSCs blends 1ml on left Achilles tendon,3 weeks 1 times,2 times;TSCs group(group B)injected by 1ml resuspending of third page of Tendon Stem Cells in density of 1*104/ml,3 weeks 1 times,2 times.PRP group(group C)were injected autologous PRP 1ml on left Achilles tendon,3weeks 1 times,2 times;model control group(group D)at the same time node were given the same amount of saline injection.After 6 weeks of intervention,animals were sacrificed and samples were collected.The tendon tissue was collected and stained.The tendon tissue was observed under the ordinary optical microscope,and the difference between the semi-quantitative scores was compared.The amount of MMP-1 was detected by ELISA,and the differences among the groups were compared.The expression of type I and ? collagen mRNA in tendon tissue was detected by qRT-PCR,and the differences among the groups were compared.Results:(1)PRP,TSC and PRP and TSC blends had different effects on the repair of Achilles tendon disease,and PRP and TSC blends was the best.A number of semi-quantitative scoring criteria score: the highest in group D,the lowest in group A.There was no significant difference between group B and group C(P>0.05).There was no significant difference between group B and group C(P>0.05).There was no significant difference between group B and group D.Compared with group D,the difference was statistically significant(P<0.05).The difference between the C group and the D group was statistically significant(P<0.05).(2)the expression of MMP-1 results by ELISA: A group compared with the blank control group,the difference was not statistically significant(P<0.05).The expression of B,C and D was statistically significant(P<0.05)compared with the blank control group.Compared with group B,C and D,the differences were statistically significant(P<0.05).There was no significant difference between group B and group C(P<0.05).There was significant difference between group B and group D(P<0.05).C and D between the two groups,the difference was statistically significant(P<0.05).(3)The expression of type I and ? collagen in fluorescence quantitative PCR showed that the expression of type I collagen mRNA in the 4 groups was significantly higher than that in the blank control group(P<0.05)The The expression of group A was the highest in group A and the lowest in group D,and the expression of B and C was between A and D.(P<0.05).There was no significant difference between group B and group C(P> 0.05).Compared with group D,there was no significant difference between group B and group B(P<0.05).The difference between the C group and the D group was statistically significant(P<0.05).The expression of type ? collagen mRNA in the four groups was significantly higher than that in the blank control group(P<0.05).The expression of group A was the highest in group A and the lowest in group D,and the expression of B and C was between A and D.(P <0.05).There was no significant difference between group B and group C(P>0.05),and there was no significant difference between group B and group B(P>0.05)(P <0.05).The difference between the C group and the D group was statistically significant(P<0.05).Conclusion:(1)In this study,the animal model of rabbit tendinopathy was induced by local injection of PGE2,and its modeling method was simple,safe and easy to control and high stability.(2)In this study,PRP was collected by Landesberg method.This method is easy to operate and promote,the collected PRP concentration was high.(3)PRP and TSC blends can restore the arrangement of tendon fibers,reduce inflammatory cell infiltration and the abnormalof tendon cells and nuclear deformation or fragmentation.(4)PRP and TSC blends can promote the expression of type I and ? collagen mRNA in tendon,and reduce the degradation mechanical properties and biomechanical properties caused by MMP-1 to collagen type I.(5)PRP and TSC blends can down-regulate the expression of MMP-1 in tendon cells and inhibit the malignant cycle of tendon cell matrix and collagen degradation,so as to delay the inflammatory response and degeneration change of tendinopathy and inhibit the progress of lesion in tendon.
