To observe the expression of PRP and TSCs combined application of rabbit Achilles tendon disease in animal models of tendon tissue structure and the content of b FGF and IGF-1 effects,an analysis preliminary of PRP combined with TSCs effect on the repair of tendon disease,PRP combined with TSCs mechanism for the treatment of tendon disease may,for the treatment of tendon disease on the basis of the relevant theory and for the clinical treatment of tendon disease and provide new ideas for the study of PRP combined with TSCs.Methods: 48 male New Zealand rabbits,after 1 weeks of feeding,were randomly divided into model group(40 rats)and control group(8).According to chemical modeling method(calcaneal attachment point distance of about 2.0cm injection of prostagland in E2 300ng/0.2ml,1 weeks / times,4 times in total)of rabbit Achilles tendon disease animal model;control group without any treatment.1 weeks after modeling,the model group again all animal were randomly divided into 5 groups,A group PRP group B group TSCs group and C group of PRP combined with TSCs group,D group and NS group,E group,model control group,8 rats in each group,blank control group F.A group was given 0.5ml PRP mold tendon around injection treatment,1 times /3 week,a total of 2 times;B group was given 0.5ml TSCs tendon injection treatment,1 times /3 week,a total of 2 times;C group was given 0.5ml PRP and TSCs suspension tendon injection treatment,1 times /3 week,a total of 2 times D;0.5ml received saline injection in the treatment of tendon around 1 times/3 weeks,a total of 2 times;E group without any treatment.The blank control group F,also no treatment.The completion of the second 3 weeks after the intervention were collected from animal specimens of rabbit Achilles tendon.The collection of Achilles tendon tissue sections after staining and observed under microscope,observe the fibers and inflammatory cell infiltration and cell arrangement etc.The collection of Achilles tendon specimens,determination of the expression of IGF-1 and b FGF content of tendon fibers in each group by ELISA method,the differences between groups were compared.Results:(1)specimens staining showed that the collagen fibers of A,B in the two groups of visible fracture,collagen fibers were wavy,rounded nuclei;collagen fibers in group C tendon specimens in a continuous,orderly,fusiform nuclei;D,E two groups had no obvious fibrous tissue.And the obvious neovascularization,more inflammatory cell infiltration.PRP,TSCs and PRP combined with TSCs on the repair of Achilles tendon repair disease in varying degrees,and the PRP combined with TSCs is better.(2)determination of IGF-1 expression: IGF-1 content from high to low were C group(4.589±0.117 ng/ml),group B(2.502±0.083 ng/ml),group A(1.384±0.028 ng/ml),group D(0.350±0.037 ng/ml),group E(0.347±0.027 ng/ml),group F(0.286±0.033 ng/ml),in C group was the highest,F group was the lowest?A,B,C three groups compared with E group,the results were statistically significant(P<0.05);group C compared with A and B between the two groups,the results were statistically significant(P<0.05);D group compared with E group,the result was not statistically significant(P> 0.05);.(3)determination of b FGF content: the content from high to low are the C group(267.415±13.251 ng/ml),B group(124.969±1.959 ng/ml),A group(62.779±3.255 ng/ml),group D(19.861±2.025 ng/ml),group E(19.251±1.114 ng/ml),group F(21.750±1.063 ng/ml),C group and high content of E group was the lowest.A,B,C three groups compared with E group,the results were statistically significant(P<0.05);group C compared with A and B between the two groups,the results were statistically significant(P<0.05);D group compared with E group,the result was not statistically significant(P> 0.05).Conclusion: PRP combined with TSCs can increase the expression levels of IGF-1 and b FGF in the recovery of tendon tissue,tendon fibers arranged,reduce the infiltration of inflammatory cells,promote the repair of tendon tissue.
