Long endurance fatigue is a kind of physiological and psychological phenomenon,which is caused by the excessive work load,the psychological factor,the bad environment and so on.And long-term single work,special environment and living conditions of the sea and other factors,clinical manifestations are reflected in the"body,nerves,feelings," three aspects.It has been reported that the incidence of long endurance fatigue is as high as 85.6%,which has gradually become the main reason for the decline of the working ability and the health condition of the long endurance,and even lead to serious accidents.At present,the long endurance fatigue is mainly in the form of clinical research at home and abroad,so it is necessary to establish a long endurance fatigue animal model,and to study the mechanism of its occurrence and development.This paper reviewed the long endurance fatigue,long endurance fatigue from the aspect of theory,the establishment of long endurance fatigue animal model in the experimental study,to explore the physiological mechanism,to provide ideas and methods for the prevention and treatment of long endurance fatigue.Theoretical research:Theory I:through summing up the domestic and international long fatigue related literature,summed up the overall fatigue fatigue performance of subjective fatigue,bad mood and cognitive decline in three areas.There are many factors that lead to long fatigue,such as long endurance environment,irregular diet and physical and psychological factors.It is precisely because of the particularity of long endurance fatigue,the current treatment of long fatigue is mainly based on recuperation and supplement,there is no animal model for long-term fatigue and effective drugs.Therefore,it is necessary to establish the animal model of long endurance fatigue.Two:the theory of modern research progress on fatigue caused by negative emotions and psychological review,widespread fatigue caused by negative emotions,the occurrence and development of multiple brain regions,mainly hippocampus,amygdala and prefrontal cortex.Its mechanism is complex,including dopamine reward mechanism,hypothalamus pituitary adrenal axis(HPA)axis disorders,5-HT metabolic disorders,GABA/GLU pathway imbalance,etc..And the treatment of traditional Chinese medicine and Western medicine treatment methods are expounded,the effect of traditional Chinese medicine is outstanding,worthy of further study and promotion.Experiment ?:the model of long endurance fatigue rat model--the establishment of the platform for shaking tableObjective:to establish a rat model of long endurance fatigue.Methods:the model of rat model was simulated by using the traditional water environment platform and floating factors.Through a large number of simulation experiments,to meet the requirements of the model to meet the requirements of the standard model,commissioned by the manufacturers for processing.Results:after many times improvement,provide specifications by us,commissioned by Beijing Keli evendata Technology Co.Ltd.production,established a model of long endurance fatigue rats-water shaking apparatus in accordance with the requirements of the platform.Experiment ?:the selection of modeling conditions for long endurance fatigue ratsObjective:to establish a suitable model for long endurance fatigue rat model.Methods:12 rats were fed and the adaptability of the site after the observation records of this group of rats in each wave is 0/min,65/min,85/min and 100/min wave when the modification time,and then statistical analysis.Results:compared with 0/min,100/min when the modification time of rats was significantly shorter,the difference is very significant(P<0.01);65/min and 85/min when the modified rat platform slightly increase and decrease,the difference was not significant(P>0.05).Compared with the 65/min,the modified time was slightly reduced at the time of 85/min,the difference was not significant(P>0.05),and the modified time was significantly shortened when the rats were treated with 100/min,the difference was very significant(P<0.01).Conclusion:the wave maker is affected by 85/min,but there is no obvious change in mood,which is the most suitable condition for the establishment of the rat model.Experiment ?:long endurance fatigue rat modelObjective:to establish a rat model of chronic fatigue and explore it.Methods:the traditional method of water environment of small standing platform rat fatigue model based on the water environment in the traditional daily stand up a small platform 10h,floating platform 2h,method for 21 days to establish a rat model of long endurance fatigue,to weight exhaustive swimming test,open field test and tail suspension test and other behavioral experiments blood lactate,creatine kinase,serum D-xylose,changes of serum corticosterone content,and water environment of small and simplestanding platform fatigue rat model comparison,comprehensive evaluation of CSC fatigue rats characteristics.Results:weight change:the rats in the normal control group were stable,and the weight of the traditional platform group was slower than that of the control group,while the weight of the floating model group was the lowest.The results showed that the exhaustion time of the floating model group was significantly shorter than that of the normal group(P<0.05),and there was no significant difference compared with the traditional group(P>0.05).The tail suspension test results show that the suspended time of the traditional platform group and the floating model group is significantly longer than that of the normal group(P<0.05).The results of field experiment showed that the modified time of rats was the longest in the floating model group,and the difference was not significant compared with the normal control group(P<0.05),and was significantly higher than that of the traditional platform group(P<0.05).The rats in the open field in the times to erect floating table model group at least,and the normal control group were significantly reduced compared with the traditional platform group,significant difference(P<0.05);the traditional platform group compared to the normal control group rearing decreased,but the difference was not significant(P<0.05).The blood lactic acid,UREA,CK results:compared with normal group,model group,floating platform blood lactic acid content was increased and the difference was significant(P<0.05);group of blood lactic acid content of traditional platform is higher than the normal group,but the difference was not significant(P>0.05).The serum UREA content,the traditional platform group compared with the normal control group had significant difference(P<0.05);floating table model group compared with the normal control group were significantly different(P<0.01),compared with the traditional platform group has no significant difference(P>0.05).The content of CK in serum was significantly higher than that in the normal control group(P<0.05).There was no significant difference between the traditional platform group and the floating model group and the normal control group(P>0.05).The serum CORT,ACTH content:two model group were significantly higher than those in normal control group(P<0.05,P<0.05),no significant difference between the model group.The serum 5-HT content:the floating platform of model group was significantly higher than the traditional platform group and normal control group(P<0.05,P<0.01),no obvious differences between the traditional platform group and normal control group(P>0.05).The serum D-xylose content:floating Taiwan model group serum D-xylose was significantly lower than the normal control group,and the difference was significant(P<0.05);no significant difference with the traditional platform group.Conclusion:the rat model of long endurance fatigue is not only full of fatigue,but also has a weak digestion and absorption ability,and the emotional reaction is affected to some extent.Experiment ?:the regulating effect of Fukang formula on the rat model of long endurance fatigueObjective:To observe the regulating effect of "Fu Kang Bu Shen prescription" on long endurance fatigue model rats.Methods:the "Tifukang Decoction" intervention on Nanjing fatigue rat model,and to the four inverse scattered and Western medicine coenzyme Q10 as control drug,to study the characteristics of its therapeutic effect on long endurance fatigue rat model and to further explore the model.Results:the"Tifukang Decoction" can significantly improve the behavior of tail suspension swimming ability,long endurance fatigue rat model of open field excitatory rats and rats,a certain regulation effect on rat blood lactic acid,UREA and 5-HT/in serum D-xylose,CORT content also play.Conclusion:the prescription of "Fu Kang Bu Shen Decoction" has a significant effect on the rat model of long endurance fatigue,and it is confirmed from the side that the pathogenesis of Chinese medicine model of long endurance fatigue model is liver depression and spleen deficiency.Experiment V:the effects of modified prescription of Fu Fu Kang on monoamine neurotransmitters in hippocampus of rats with chronic fatigue modelObjective:To investigate the effect of Fukang Decoction on monoamine neurotransmitters in hippocampus of rats with chronic fatigue and to explore the mechanism of action.Methods:the hippocampus tissue of four rats were taken and the contents of 5-HT,DA and NE were determined by hplc.Results:the floating platform in hippocampus of model group rats 5-HT content was significantly lower than control group,significant difference(P<0.05),each group of rat hippocampal 5-HT content compared with the model group increased the floating platform;among the other groups,there is no significant difference between treatment group and control group(P>0.05).There was no significant difference between DA group and NE group than that in the control group and the control group(P>0.05).Conclusion:changes in fatigue rats have depression,but did not reach to the diagnosis of depression,"Tifukang Decoction" to regulate mood long endurance fatigue rats may be related to its effect of improving the content of 5-HT in hippocampus.SummaryThe conclusion is as follows:first,the long endurance fatigue rat model has obvious negative emotions of depression.Two,the body of the rehabilitation and rehabilitation of the long fatigue rat model of fatigue and negative emotions have a significant role in improving.Three,long endurance fatigue rats have depression,but did not reach to the diagnosis of depression,"Tifukang Decoction" to regulate mood long endurance fatigue rats may play a therapeutic effect by adjusting the content of 5-HT in hippocampus.
