| Objective:1.To study the composition principles of prescriptions in traditional Chinese medicine for acute intracerebral hemorrhag(AICH)through the literature excavation based on Traditional Chinese Medicine Inheritance Support System(TCMISS,V2.5),hoping to provide reference and basis for the treatment of cerebral hemorrhage with traditional Chinese medicine.2.To investigate the protective effect of Ditantongyu Decoction and the change of expression of IRAK-M mRNA in Kupffer cells(KCs)caused by hepatic stress injury in rats with acute cerebral hemorrhage by experimental study,which could provide experimental basis for the clinical application of Ditantongyu decoction.Method:1.Literature mining:Selecting the prescriptions in traditional Chinese medicine for ACH in the literatures of China National Knowledge Internet(CNKI)from 1991 to 2016,using unsupervised data mining method provide by TCMISS,digging out the high-frequency drugs,couplet medicines,core combinations,new prescriptions and so on,The medicine compatibility rule,which can summarize the composition principles of these treatment of acute intracerebral hemorrhag.2.Experimental study:Cerebral hemorrhage was induced by injecting collagenase Ⅶ into the caudate nucleus of rats,healthy SD rats were randomly divided into 3groups:blank control group,cerebral hemorrhage group and Ditan Tongyu group(the rtubarb,bravery south start,stone calamus,cattail pollen,qseudo-ginseng,leech).collectting the serum frozen spare parts-20 ℃.KCs were obtained through density gradient centrifugation and collected at 6,12,and 24h after drug administration.Expression of IRAK-M mRNA in KCs tested by using polymerase chain reaction(PCR)method;the changes of nuclear factor-κB(NF-κB)was detected by Western blot;the levels of tumor necrosis factor-α(TNF-α)were determined by Elisa and the cell apoptosis were analyzed by flow cytometry.Result:1.Literature mining:collect 312 literature articles,which meet the standards and get 318 prescriptions,involving 206 kinds of traditional Chinese medicine.The drugs of top ten frequency are Rheum officinale,Qseudo-ginseng,Stone-calamus,Leech,Salvia-miltiorrhiza,U-ncariae,Achyranthes,Bile-arisaema,Gastrodia-tuer,Ligusticum-wallichii.Rheum-officinale was used with the highest frequency(199 times);7 core combinations and 7 new prescriptions were discovered.The core drug is Rheum-officinale and the common drugs of prescription which contains Rheum-officinale and Qseudo-ginseng(101 times).2.Experimental study:Ditantongyu group decreased in blank control group,but it elevated significantly compared with model group(P<0.05);Compared with cerebral hemorrhage group,the levels of NF-κB and TNF-α in KCs in Ditan Tongyu Decoction group decreased significantly(P<0.05,P<0.01);There were many apoptotic cells showed in cerebral hemorrhage group and apoptosis rate of KCs in Ditan Tongyu Decoction remarkably lower than that of model group(P<0.05).Conclusion:1.With the medication rules for acute intracerebral hemorrhag were analyzed by data mining,Rhubarb is the most widely used;The main combination of rhubarb and pseudo-ginseng;The main drug species are the kind of heat-clearing and detoxifying,phledm-heat and fu-organexcess,promoting blood circulation by removing blood stasis.The application of TCMISS in the traditional treatment of acute intracerebral hemorrhag has important guiding significance.2.Ditantongyu decoction could increase expression of IRAK-M mRNA in KCs,block activation of NF-κB and release of TNF-aavailably and inhibit apoptosis of KCs,which is an important way for preventing and treating hepatic stress injury caused by ACH. |
